温阳活血方对体外鼠肾成纤维细胞FN、LN表达的影响

摘　　要：目的:探讨纤维粘连蛋白(FN)、层粘连蛋白(LN)在体外鼠肾成纤维细胞的表达及温阳活血方的干预作用.方法:体外培养健康SD大鼠肾成纤维细胞,随机分为6组:假手术组、模型组、温阳活血方组、肉苁蓉组、桃仁组、福辛普利组.检测药物血清干预细胞72h培养上清中FN、LN浓度.结果:(1)模型组FN、LN浓度显著高于假手术组,各药物组对FN、LN均有抑制作用.(2)对FN的抑制作用除福辛普利各剂量组无显著差异外,其他同一药物的不同剂量间呈现一定的剂量依赖性;温阳活血方大剂量组显著低于桃仁和肉苁蓉大剂量组;各药物中剂量组间无显著差异;各药物小剂量组,温阳活血组低于桃仁组,高于肉苁蓉组,差异有显著性.(3)对LN的抑制作用除桃仁各剂量组间无差异外,其他同一药物的不同剂量间呈现一定的剂量依赖性;各药物大剂量组间差异无显著性;各药物中、小剂量组,温阳活血组小于福辛普利组和桃仁组,差异有显著性,与肉苁蓉组比较差异无显著性.结论:温阳活血方可能通过抑制成纤维细胞分泌FN、LN,进而减少细胞外基质(ECM)堆积,从而对肾间质纤维化起治疗作用.Objective：To investigate Wenyang Huoxue recipe that could induce the expression of fibronectin （FN） and laminin （LN） by rat renal fibroblasts in vitro culture. Methods：Health SD rats renal fibroblasts were cultured in vitro and randomized into 6 groups： sham operated group, model group, Wenyang Huoxue recipe group, herba cistanches group, semen persicae group, fosinopril group. The level of FN and LN in culture solution was detected after 72 hours of medicamentous blood serum effect. Resuhs：（1）The level of FN and LN was significantly higher in model group than in sham operated group. Ever medicamentous groups could reduce the expression of FN and LN. （2）To the extression of FN, the different dosage of same agent showed invariable dose dependent except fosinopril group. Wenyang Huoxue recipe large dose group could significantly decrease the expression of FN than herba cistanches large dose group and semen persicae large dose group. Ever middle dose groups hadn＇ t markedly difference to decrease the expression of FN. In low dose group, Wenyang Huoxue recipe group could significantly decrease the expression of FN than semen persicae group. However, herba cistanches low dose group was stronger than Wenyang Huoxue recipe group. （3）To the extression of LN, the different dosage of same agent showed invariable dose dependent except semen persicae group. Ever large dose groups hadn＇ t markedly difference to decrease the expression of LN. In middle group and low group, Wenyang Huoxue recipe groups were thinner than semen persicae groups and fosinopril groups except herba cistanches groups. Conclusion：Wenyang Huoxue recipe could markedly inhibit fibroblasts to excrete FN and LN through herba cistanches and semen persicae. Furthermore, Wenyang Huoxuc recipe could decreace extracellular matrix to alleviate renal interstitial fibrosis.
