TSHR及活性多肽对BALB/c小鼠甲状腺的影响

摘  要:目的:探讨促甲状腺激素受体(TSHR)及活性多肽对BALB/c小鼠甲状腺的影响。方法:对BALB/c小鼠腹腔注射血蓝蛋白耦联的人TSHR(hTSHRaa352~366)和从豚鼠脂肪细胞提取的TSHR(gTSHR)进行免疫,每2周免疫1次共6次,每次免疫前2d取血,检测血清T4、促甲状腺激素受体抗体(TRAb)水平。12周后处死小鼠,取甲状腺做病理组织学检查。结果:血清T4水平hTSHRaa352~366组从第6周开始下降(P<0.01),而且一直维持到12周,血清TRAb水平明显升高(P<0.01),甲状腺组织显示滤泡上皮细胞扁平、核缺失、皱缩、胶质浓缩等甲状腺功能减退病理改变;gTSHR组血清T4水平在第6周已下降,而12周时有的下降、有的则升高,无统计学意义,血清TRAb水平升高(P<0.01),甲状腺组织出现不均一的病理表现。结论:hTSHRaa352~366刺激BALB/c小鼠产生TRAb和促甲状腺激素受体抑制抗体(TBAb),抑制促甲状腺激素(TSH)与TSHR结合,降低了甲状腺激素T4水平;而gTSHR刺激BALB/c小鼠同时产生TRAb、促甲状腺激素受体刺激抗体(TSAb)和TBAb,引起甲状腺功能亢进或甲状腺功能减退。Objective: To study the influence of thyrotropin receptor (TSHR) and its active polypetide fragment on thyriod in BALB/c mice. Methods: BALB/c mice were injected peritoneally with TSHRaa352-366-KLH ( hemocyanin from keyhole limpets) and guinea pig TSHR(gTSHR) every 2 weed, respectively .The levels of thyroid hormones and TSHR antibodies were observed .The animals were killed on the week 12, and the thyroid were taken, and the pathological changes of thyroid tissue were observed. Results: In the group injected with TSHRaa352-366-KLH, the serum levels of T4 decreased from the 6th week to 12th week (P〈0.01) and levels of thyrotropin receptor antibody (TRAb) increased (P〈0.01). The thyroid follicles showed some pathological changes such as flatted epithelia, shrinkage or bacilliform karyon in some epithelia and condensed colliod matreial et al like hypothyroidism . In gTSHR group, the serum levels of T4 had decreased at 6 week, but some animals falled continue and some animals rised at 12 week, they had no significance of statistics. Serum levels of TRAb increased (P〈0.01) compared with the date in same group at day 0, and the thyroid follice showed some herterogeneous pathological changes. Conclusion: hTSHRaa 352-366-KLH as an activated antigen may induce TRAb and thyrotropin receptor blocking antibody (TBAb), block the binding of TSH and TSHR in mice and inhibit the secretion of T4, gTSHR may induce thyrotropin receptor stimulating antibody (TSAb) and TBAb in mice and lead to hypothyrodism or hyperthyroidism.
