摘  要:目的观察大鼠放射性肺损伤模型中血浆转化生长因子β1(TGF-β1)的变化以及灯盏细辛和曲尼斯特对其的影响。方法90只Wistar大鼠随机分为4组,空白组15只、单照组25只、曲尼斯特组25只、灯盏细辛组25只。单照组和两药物组用直线加速器产生的6 MV X射线照射右全胸,单次20 Gy。曲尼斯特组于照射前2周、灯盏细辛组于照射后开始每天灌胃给药,给药剂量曲尼斯特溶液50 mg/kg、灯盏细辛注射液50 mg/kg,持续到照射后2个月。空白组与单照组均灌胃相等体积的生理盐水。照射后每月取大鼠静脉血浆标本行TGF-β1检测,观察其变化情况。结果单照组血浆TGF-β1含量在照射后1个月开始增加,高峰在照射后第2个月,以后逐渐回落至一高水平。两药物组血浆TGF-β1含量2个月时明显低于单照组,但停药后仍出现高峰,高峰延至照射后第3个月。两药物组与单照组的TGF-β1含量均高于空白组,与肺损伤的程度一致。结论灯盏细辛和曲尼斯特能明显降低照射后血浆TGF-β1的含量,延长给药时间是否能增加疗效值得研究。Objective It is to observe the change of transforming growth factor β1 (TGF- β1 ) in rats with radiation in- duced lung injury and the influence of Tranilast and fleabane on it. Methods Ninty Wistar rats were randomly divided into four groups: blank group, only irradiated group, Tranilast group and fleabane group. Only irradiated group, Tranilast group and fleabane group were irradiated with X - rays at a dose of 20 Gy thoracically. Tranilast group started intragastric adminis- tration Tranilast at a dose of 50 mg/kg per day before irradiation for two weeks and fleabane group started intragastric admin- istration fleabane at a dose of 50 mg/kg per day after irradiation for continuing to two months after irradiation. Blank group and only irradiated group were given the same volume normal sodium intragastric administration. Plasma TGF- β1 level was measured monthly after irradiation. Results Plasma TGF - β1 level in only irradiated group showed an increase from one month after radiation, peaking at two months and declining at a high level after two months. In the two drug groups, plasma TGF- β1 contents were significantly lower than that in only irradiated group but peaking at three months after irradiation while stopping drugs. Plasma TGF- β1 contents in only irradiated group, Tranilast group and fleabane group were all higher than that in blank group, it was conformity with the degree of lung injury. Conclusion Fleabane and Tranilast can obviously lowered the contents of plasma TGF- β1 after radiation. Prolonging administer time can or cannot increase curative effect is worth to further study.
