摘 要:在传统的放射免疫分析的基础上,以磁性微粒子为二抗分离剂和固相一抗为结合反应剂,建立了两种快速、超灵敏、简便的血清及其它样品中雌三醇含量的分析方法。雌三醇测定的曲线范围0.3~30μg/L;磁性微粒子二抗分离法批内和批间相对标准偏差分别为4.3%和8.7%;回收率为104%~114%;健全性为0.9989。磁性微粒子固相一抗法批内和批间相对标准偏差分别为4.4%和7.6%;回收率为96%~115%;健全性为0.9986。两种方法相比,磁性微粒子固相一抗法具有更为简便、快捷的优点。两种磁性微粒子的放射免疫分析不仅缩短了反应时间,其准确性和精密度也均满足临床及环境研究中的检验要求,且灵敏度较现有方法提高了一个数量级。In the present work, using the magnetic microparticle, two ultra-sensitive and simple methods based on the traditional radio-immunoassay for the determination of estriol (E3) in serum were developed. Magnetic microparticles were used as the second-antibody separation agent and the first-antibody solid-surface blocked agent. Under the selected conditions, the measurement calibration curve for E3 was in the range of 0. 3 -30μg/L. The intra- and inter- RSD of second-antibody method was 4.3% and 8.7% respectively. The recovery was 104% - 114% , and the soundness coefficient was 0. 9989 ; The intra- and inter- RSD of firstantibody method was 4.4% and 7.6% respectively. The recovery rate was 96% -115% , and the soundness coefficient was 0. 9986. By these two methods using magnetic microparticle can be shortened. The accuracy and precision are well enough for clinical and environmental analysis. Compared these two methods, magnetic microparticle using as the first-antibody was more simple and faster.
