摘 要:目的探讨人主动脉-性腺-中肾(AGM)区基质细胞对脐血CD34+细胞形成高增殖潜能集落形成单位(HPP-CFU)能力的支持作用。方法采用免疫磁珠方法分离人脐血CD34+细胞,接种于底层已制备好人AGM区基质细胞S1~S5饲养层的24孔板中,同时设无饲养层组及取自同期胚胎的躯干成纤维细胞hFT为对照,体外培养28天,每7天收获细胞行造血细胞集落培养。结果5株人AGM区基质细胞hAGMS1~S5对HPP-CFU均有不同程度的扩增及维持作用,HPP-CFU在培养14天达到峰值,与无饲养层组、hFT组比较差异有统计学意义(P<0.05)。hAGMS1~S5的造血支持作用组间比较差异亦有统计学意义(P<0.05),其中hAGMS3、S4优于S1、S2及S5。结论人AGM区基质细胞S1~S5对脐血CD34+细胞形成HPP-CFU的能力具有支持作用,特别是hAGNS3、S4两株细胞具有更好的造血干性维持作用,为胚胎早期造血发生机制研究提供了重要的模式细胞和基础资料。Objective To explore the supportive effects of stromal cells derived from huaman aotra-gonad-mesonephros (AGM) region on the ability of umbilical cord blood CD34 ^+ cells to form HPP-CFU. Methods Stromal cells derived form human AGM region (hAGMS1 - S5) and fetal trunk fibroblasts (hFT) were cultured on 24-well plates as feeder cells. CD34^+ cells were positively selected from human umbilical cord blood through immunomagnetic beads selection method, seeded into 24- well plates, and co-cultured for 28d. During co-culture, hematopoietic cells were collected every week for colony analysis in methylcellulose semisolid medium. Results AGM stromal cells sigmificantly supported the expansion of HPP-CFU, when compared with hFT and controls without feeder cells. HPP-CFU got to peak value at day 14 of co-culture. The supportive effects of human AGM S1 - S5 were significantly different, hAGM S3 and S4 were better than hAGM S1, S2, and S5. Conclusion Human AGM stromal cells S1 - S5 could support umbilical cord blood CD34 ^+ cells to form HPP-CFU, and hAGM S3, S4 cell had better effects of maintaining HSC activity, which would provide model cells and basic data for researches on hematopoiesis mechanism.
