摘 要:目的探讨苦参合剂对隐孢子虫感染BALB/c小鼠免疫功能的保护机制。方法建立隐孢子虫病(CPS)小鼠模型,给小鼠苦参合剂,4周后剖杀,进行脾细胞的分离和培养,采用氚-胸腺嘧啶核苷(3H-TdR)掺入技术体外免疫实验法和四唑盐比色试验(MTT整体免疫实验)检测小鼠T淋巴细胞的转化增殖情况,实验设有单纯模型组、PHA阳性对照组及正常对照组。结果3H-TdR掺入技术体外免疫实验中,500、250、125、62.5和31.25μg/ml剂量组的β射线放射活性(cpm值)均显著高于正常小鼠和CPS模型小鼠不给药对照组(P<0.05或P<0.01),以62.5μg/ml浓度组最强;MTT法整体实验中,苦参合剂(0.2 ml/只)用药组的吸光度(A)值与正常对照组相比显著升高(P<0.05)。结论苦参合剂能刺激T淋巴细胞的转化,增强T淋巴细胞的增殖,提高机体的细胞免疫功能。Objective To study the protective mechanism of the radix sophorae flavescentis(RSF) mixture on cellular immunity of BALB/c mice against Cryptosporidiosis(CPS). Methods The mouse model of CPS was established. Mice were administered RSF mixture for 4 weeks, and the spleen cells were segregated and cultivated. The multiplication and transformation of T lympholeukocyte were valued by lymphocyte transformation test including ^3 H-TdR incorporation and MTT colorimetric method. Results In the vitro immunity experiment of ^3 H-TdR incorporation, the radiate activity of B-ray (cpm fig) in experimental groups with 500, 250, 125, 62.5 and 31.25 μg/ml were higher than the control groups (normal and CPS model with no administration) (P〈0.01 or P〈0. 05), especially the 62.5 μg/ml group. The absorbance (A) of RSF mixture group was higher than the normal control group(P〈0.05) by MTT assay. Conclusion The RSF mixture can stimulate and strengthen the transformation and multiplication of T lympholeukocyte, elevate the host' s cellular immunity.
