摘  要:目的:考察桂竹糖芥苷对血管平滑肌细胞增殖的影响。方法:应用80%乙醇回流提取,正己烷萃取,MCI柱层析甲醇洗脱获得桂竹糖芥苷。采用体外培养的血管平滑肌细胞,检测细胞增殖、细胞内钙和c-myc基因的mRNA表达,并与哇巴因、地高辛进行比较。结果:(1)桂竹糖芥苷浓度在10-5mg·mL-1时具有促血管平滑肌细胞增殖作用;哇巴因和地高辛浓度分别在10-14和10-9mg·mL-1时具有促血管平滑肌细胞增殖作用,最大增殖率分别为49.08%和51.05%。(2)桂竹糖芥苷浓度在10-3mg·mL-1时可暂短升高细胞内钙;哇巴因浓度在10-4mg·mL-1时可持久升高细胞内钙。(3)桂竹糖芥苷浓度在10-8mg·mL-1时对c-myc mRNA表达没有影响;哇巴因和地高辛浓度在10-12mg.mL-1时可增加c-myc mRNA表达。结论:与哇巴因和地高辛相比,桂竹糖芥苷对血管平滑肌细胞增殖的影响较小。AIM: To investigate the effects of erysimosides on proliferation in vascular smooth muscle cells. METHODS: Erysimosides were obtained by extraction with 80% ethanol and isolation with MIC column. Effects of erysimosides on proliferation, intracellular free calcium and c-myc mRNA expression in vascular smooth muscle cells were observed and compared with those of ouabain and digoxin. RESULTS: (1) Erysimosides at the concentration of 10^-5 mg· mL^-1 presented the effect of stimulating proliferation; Ouabain and digoxin presented the effect of stimulating proliferation at the concentration of 10^-14 mg· mL^-1 and 10^-9 mg·mL^-1, respectively. (2) Erysimosides at the concentration of 10^-3 mg·mL^-1 appeared a tentative increase of intracellular free calcium; Ouabain at the concentration of 10^-4 mg · mL^-1 appeared a permanent increase of intracellular free calcium. (3) Erysimosides at the concentration of 10-8 mg·mL^-1 had no effect on c-myc mRNA expression; an increase of c-myc mRNA expression was observed by ouabain and digoxin at the concentration of 10^-12 mg · mL^- 1 . CONCLUSION: Compared with ouabain and digoxin, the effects of erysimosides on proliferation in vascular smooth muscle cells is small.
