摘 要:目的建立猪动脉粥样硬化(AS)模型,探讨该模型中LOX-1表达的变化。方法将10头广西巴马小型猪分别喂正常(CD)和高脂、高糖、高胆固醇(HFSCD)饲料,每月末测定血脂水平。第5个月末处死动物,采用苏丹Ⅳ检测动脉粥样斑块病变;逆转录-聚合酶链反应(RT-PCR)、Western Blot印迹分别检测主动脉组织中LOX-1的mRNA和蛋白质表达。结果与CD组比较,HFSCD组血浆甘油三酯、高密度脂蛋白胆固醇(HDL—C)和总胆固醇水平异常升高;动脉有明显的脂质条纹及斑块;动脉粥样硬化小型猪主动脉组织的LOX-1的mRNA及蛋白质的表达均上调(P<0.05)。结论高脂、高糖、高胆固醇饲料喂养小型猪可建立AS动物模型;AS小型猪主动脉组织的LOX-1表达上调。 Objective To study the expressions of lectin-like oxidized low-density lipoprotein receptor-1 in atherosclerotic mini swine and provide a new mechanism for investigating the pathogenesis of atherosclerosis. Methods Ten Guangxi Bama minipigs were randomized into two groups:control(CD)group(n=5),fed with normal control diet),high-fat/high-sucrose/high-cholesterol diet HFSCD group (n=5,fed with the high-fat/high-sucrose/high-cholesterol diet). Plasma total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C)and triglyceride (TG)were examined at the end of every month. The expressions of LOX-1 mRNA and protein in aorta tissue were detected by reverse transcriptase-polymerase chain reaction(RT-PCR) and Western Blot. Results At the end of five months,plasma TC,HDL-C and TG in HFSCD mini swine were increased. There were obvious fatty streak and atherosclerotic plaque in mini swine aorta. The expression of LOX-1 was upregulated in HFSCD mini swine aorta tissue. Conclusion HFSCD may induce atheroselerosis and the expression ofLOX-1. Upregulating LOX-1 expression may promote atherosclerosis.
