摘 要:目的探讨微波辐射对大鼠大脑皮层和海马突触囊泡相关蛋白如突触素I(synapsinI),囊泡相关膜蛋白-2(vesicle-associated membrane protein-2,VAMP-2)和突触融合蛋白(syntaxin)以及突触囊泡蛋白(synaptophysin)表达的影响。方法采用30mW/cm2微波辐射Wistar雄性大鼠,于辐射后6h,1,3和7d取材,通过免疫蛋白印迹检测大脑皮层和海马突触体突触素I,VAMP-2,突触融合蛋白和突触囊泡蛋白表达的改变;采用免疫共沉淀检测VAMP-2和突触融合蛋白相互作用的改变。结果30mW/cm2微波辐射后皮层突触素I于辐射后3d表达减少(P<0.01);海马突触素I表达1d增加,3d减少,7d又增加的波动(P<0.01)。皮层和海马突触囊泡蛋白于辐射后7d表达增加(P<0.01),VAMP-2和突触融合蛋白于辐射后7d内表达均降低。VAMP-2和突触融合蛋白相互作用在皮层于辐射后3d减弱,而海马于辐射后7d内明显减弱(P<0.01)。结论微波辐射可引起大脑皮层和海马突触囊泡蛋白表达异常,进而影响突触传递功能。Objective To investigate the effect of microwave radiation on expressions of synaptic associated proteins: synapsin I, VAMP-2, syntaxin and synaptophysin. Methods 25 Wistar rats were exposed to microwave which average power density was 30 mW/cm^2. Western blot was used to study the change of the expressions of synapsin I, VAMP-2, syntaxin and synaptophysin of synaptosome in cerebral cortex and hippocampus, and immune co - precipitation was used to study the interaction of VAMP-2 and syntaxin. Results It resulted in the decrease of synapsin I at after 3-day radiation in cerebral cortex (P 〈 0.01 ), but increase at 1 d, decrease at 3 d and increase at 7 d in hippocampus (P 〈 0.01 ) ; the increase of synaptophysin at 1 -- 7 d and decrease of VAMP-2 and syntaxin at 6 h-- 7 d after radiation in cerebral cortex and h/ppocampus (P 〈 0.01 ) ; the decrease of the interaction of VAMP-2 and syntaxin at 3 d in cerebral cortex and at 6 h-- 7 d in hippocampus after expo- sure (P 〈 0.01 ). Conclusion Microwave radiation can induce the abnormity of expressions of synaptic associated proteins in cerebral cortex, and hipoccampus then induce the disorder of synaptic transmission.
