摘  要:目的通过低密度脂蛋白(LDL)的刺激,观察LDL对体外培养的大鼠肾小球系膜细胞(MCs)表型转化的诱导作用。方法采用不同浓度的LDL(0,25,50,100,150μg/ml)刺激体外培养的大鼠肾小球系膜细胞,以四甲基偶氮噻唑蓝(MTT)比色法检测细胞增殖,透射电镜观察系膜细胞的形态学变化,免疫细胞化学染色及蛋白印迹法(Western blot)检测α-平滑肌肌动蛋白(α-SMA)、结缔组织生长因子(CTGF)的表达。结果在LDL作用下,倒置显微镜下细胞变为长梭形,透射电镜下细胞表面微绒毛减少。免疫细胞化学染色及Westernblot显示α-SMA、CTGF表达增强(P<0.05)。结论在LDL刺激下,系膜细胞可以发生表型转化并可能与肾小球硬化有关。Objective To study the influence of low density lipoprotein on phenotype change of the cultured rat glomerular mesangial cells(MCs). Methods The rat glomenllar mesangial cells were cultured for seven days in low density lipoprorein(0, 25, 50,100,150vg/ml). MCs proliferation was measured under MTT assay. Morphological changes were observed by electronic microcopy. α- SMA,CTGF were examined by immunohistochemistry stain and Western blot. Results Glomerular mesangial cells cultured in low density lipoprotein showed obvious morphlogic changes, including elongated shape and the appearance of muscle actin. The expression amount of α - SMA. CTGF enhanced by using immunohistochemistry stain and Western blot. Conclusion Low density lipoprotein can lead to the phenotypic change of glomenllar mesangial cells and have much something with fibrosis of the kidney.
