摘 要:在pH7.5磷酸盐缓冲介质中,硝酸盐还原酶可还原硝酸盐为亚硝酸盐,Cu2+的存在能够抑制此反应的进行,并且与还原程度在一定范围内成线性关系。通过间接测定反应生成的NO2-浓度能够确定Cu2+的浓度,与自制的光反射传感器联用,建立了水中铜的现场快速定量检测方法。讨论了显色剂用量、还原酶用量、反应时间、温度等因素的影响;方法的线性范围为0.01~2.00mg/L,检出限0.005mg/L,相对标准偏差4.0%,加标回收率在96.2%~104.2%之间。本方法可以非常方便地应用于环境水体中铜的现场快速定量测定。A quantitative determination method of the copper(Ⅱ) in water based on an enzyme inhibition reaction was developed. Nitrate was deacidized to nitrite with nitrate reductase in phosphate buffer solution(pH 7. 5). The reaction was inhibited when copper(Ⅱ) existed. The concentration of Cu^2+ was known by the indirect determination of NO2^- with a refelectmeter. Effect of temperature, time of reaction,dosage of developer and nitrate reductase on the determination of copper was studied. The linear range was 0.01--2.00 mg/L,the detection limit was 0.05 mg/L and the relative standard deviation was 4.0% for Cu^2+ (n=11). The recoveries in water samples were 96.2%-104. 2%. It was shown that the method is reliable for the test of copper in-situ analysis.
