摘 要:【目的】观察消瘕丸对大鼠乳腺增生(Mammary Gland Hyperplasia:MGH)动物模型的影响,阐明其作用机理。【方法】以外源性激素诱导大鼠建立乳腺增生病模型,观察其对乳腺的组织形态、乳头大小的影响及血清中超氧化物歧化酶(superoxide dismutase,SOD)活性和丙二醛(MDA)的含量变化;采用免疫组化SP法检测大鼠乳腺组织雌激素受体(estrone receptor,ER)、孕激素受体(progesterone receptor,PR)的表达。【结果】与模型组相比,消瘕丸治疗组能抑制乳腺组织增生,以高剂量组效果最显著,血清中SOD活性升高(P<0.01)、MDA含量降低(P<0.05);ER、PR的表达明显减弱,(P<0.01,P<0.05)。【结论】消瘕丸治疗乳腺增生的机理可能与其抑制ER、PR的表达,降低MDA含量增加SOD的活性有关。[ Objective] To observe the effect and pharmacological mechanism of XiaoJia Wan pill in treating MGH in rats. [Methods] Model of MGH induced by sex hormone was established in Wistar rats. Pathologic changes of breast tissue and the changes of activities of SOD, MDA in serum were observed. And immunohistechemical SP staining was performed to test the expressions of RP protein and EP protein in mammary hyperplasia in rats. [Results] Compared with model groups, SOD was markedly raised and MDA was significantly lower in XiaoJia Wan groups, simultaneously, the pathologic changes in treated group were eontrolled. The expresstions of EP and RP were reduced significantly. [ Conclusions] The mechanism of the obvious curative effect XiaoJiaWan MGH may be due to the reduce of EP, RP and MDA and increase of SOD in MGH in rats.
