摘 要:目的研究2,3,7,8-四氯二苯并二噁英(2,3,7,8-tetrachlorodibenzo--pdioxin,TCDD)对大鼠血清中氧化应激指标的影响。方法清洁级Wistar雄性大鼠32只,体重(100±10)g,随机分成4组,连续经口染毒TCDD 90 d,剂量分别为2.5、25、250 ng/kg,对照组给予二甲基亚砜(DMSO),测定SOD、CuZn-SOD、Mn-SOD、GSH-Px、GST活性及MDA的含量。结果TCDD染毒90 d后,与对照组相比,各染毒组雄性大鼠血清中MDA含量明显增高(P<0.05);各染毒组总SOD、CuZn-SOD、MnSOD、GSH-Px活力均有所下降,其中除CuZn-SOD外,与对照组相比差异均有统计学意义(P<0.05)。同时各染毒组GST活力显著增加,且各组间差异有统计学意义(P<0.05)。结论亚慢性染毒引起MDA含量增加,大鼠血清中抗氧化指标出现异常改变,提示体内出现氧化与抗氧化系统失衡。Objective To study the effect of 2,3,7,8-tetrachloredibenzo-p-dioxin (TCDD) on the levels of antioxidative system of male Wistar rats. Method 32 healthy male Wistar rats were randomly divided into four groups: three were exposed groups (respectively exposed to 2.5 ng/kg, 25 ng/kg and 250ng/kg TCDD) and a control group. The rats were orally given TCDD or DMSO, respectively for 90 days, then the rats as killed and the activities of SOD, CuZn-SOD, Mn-SOD, GSH-Px, GST and the levels of MDA in the serum of the rats were detected. Result The MDA level in serum of exposed rats was significantly increased ( P 〈 0.05) ; while the activities of SOD, CuZn-SOD. Mn-SOD and GSH-Px were decreased in all TCDD-treated group (P 〈 0.05) except CuZn-SOD, while, the serum activity of GST significantly increased (P 〈 0.05). Conclusion The subchronic exposure to TCDD could damage serum antioxidative activity, induce the increase of MDA and the abnormalities of antioxidative indices in serum of Wistar rats. It is suggested that there is some unbalance between oxidation stress and antioxidafive system.
