平阳霉素磁性脂质体对人粘液表皮样癌的体内抗瘤作用研究

摘　　要：以荷人粘液表皮样癌的裸鼠作为动物模型，采用局部给药方式，同时在肿瘤部位放置０．２３Ｔ恒定强磁场对肿瘤进行磁导向化疗。实验结果显示，平阳霉素磁性脂质体对人粘液表皮样癌的移植瘤有显著抑制作用，抑瘤率为８１．１％，显著高于平霉素脂质体（６０．９％）和游离平阳霉素（４５．５％），而且其对正常组织（主要是肺脏）的毒副作用明显减轻。To study the antitumor effects of pingyangmycin magnetic liposome on human mucoepidermoid carcinoma, MEC-1 cells were implanted into 20 nude mice subcutaneously. The mice were randomly divided by 5 in each group and treated with local administration of pingyangmycin-magnetic-liposome (PYM-M-Lip),pingyangmycin-liposome (PYM-Lip), pingyangmycin (PYM) and normal saline (control) respectively. The tumor size and body weight were measured regularly, the tumor growth inhibition and the body weight loss were calculated. Histological changes were studied with light and electron microscopes. Stronger tumor growth inhibition in PYM-M-Lip treated mice (81.1%) was obtained than in PYM-Lip (60.9%),PYM(45.5%) and control (0);less body weight loss was observed in the drug treated mice (2.3%～3.2%) than in the control(14.2%). Necrosis of the tumor tissue and vesicles containing magnetic granules in the tumor cells were found in PYM-M-Lip treated mice. The results indicate that PYM-M-Lip is stronger in the inhibition of MEC-1 cell growth in nude mice with weaker side effects than PYM-Lip or PYM.
