摘  要:目的通过骨组织形态计量学方法观察和比较中药补骨胶囊与西药羟乙膦酸钠对去睾丸(ORX)大鼠骨代谢的影响。方法40只3月半龄SD雄性大鼠,随机分成年龄对照组、ORX组、ORX分别加羟乙膦酸钠和补骨胶囊组。前两组每日生理盐水,羟乙膦酸钠组药物按36mg/kg.d,补骨胶囊用药组按5mL/kg.d(330g/kg),共灌胃90d。实验结束,取胫骨近心端进行不脱钙骨制片进行骨组织形态计量学分析。结果ORX组骨小梁面积百分率(%Tb.Ar)比对照组降低(-50.7%,P<0.05),代表骨形成和骨吸收的参数值均显著增加。与ORX组相比,羟乙膦酸钠组%Tb.Ar增加(+86.1%,P<0.05),补骨胶囊组%Tb.Ar增加(+54.0%,P<0.05),代表骨形成和骨吸收的参数值均降低,两组间差异无显著性。结论羟乙膦酸钠和补骨胶囊均能防止ORX大鼠造成的骨量丢失,两者的效果相当,选用中药副作用小。Objective To study the skeletal effects of Bugu capsule and etidronate in orchiectomized(ORX) rats by bone histomorphometry. Methods Forty three-and-half-month-old Sprague Dawley male rats were randomly divided into four groups: aging control(sham), orchiectomized group(ORX), ORX with etidronate group and ORX with Bugu capsule group. Group Sham and ORX with 0.9% Nacl daily, the other two groups were injected through mouth with etidronate 36 mg/kg· d and Bugu capsule 5 mL/kg per day (330 g/kg) respectively. After 90 days, all rats were sacrificed and their proximal tibial(PTM) were processed into undecalcified sections for histomorphometric analysis. Results Compared with sham control, ORX group manifested reduction of bone mass (percent trabecular area % Tb. Ar -50.7 %, P 〈 0.05). Indexes of bone formation and bone resorption rate both increased markedly. %Tb. Ar increased by 54.0% ( P 〈 0.05) in Bugu group and by 86.1% ( P 〈 0.05) in etidronate treatment group compared with that of ORX. Dynamic data showed that bone formation and resorption rate both reduced, but with no significance between two groups. Conclusions The results indicate that the Bugu capsule could prevent the bone loss in ORX rats as well as etidronate.
