摘  要:用荧光探针Fura-2及BCECF结合计算机图象处理技术,观察氨氯地平对缺氧和复氧损伤心肌细胞内pH值和Ca2+浓度的影响。发现缺氧和复氧损伤细胞内游离钙离子的浓度明显上升,pH值下降;氨氯地平能显著地降低缺氧损伤的心肌细胞内的游离Ca2+浓度,并抑制细胞酸化;但对缺氧损伤后再复氧的心肌细胞内游离Ca2+的浓度和pH值均无显著影响。Fluorescent probes combined with digital image processing technique were used to study the effect of amlodipine on intracellular pH and calcium concentration in hypoxic and reoxygenated cardiac myocytes. The results indicated that the calcium concentration significantly increased while the pH decrased during hypoxia and reoxygenation. Amlodipine could significantly reduce free calcium concentration and inhibit intracellular acidosis during hypoxia. However, it had no effect on variations of both pH and calcium concentration induced by reoxygenation.
