免疫力素对小鼠L6565逆转录病毒的作用

摘　　要：目的:观察免疫力素对小鼠L6565逆转录病毒模型的治疗作用,以探讨其治疗艾滋病的可能性。方法:用小鼠L6565病毒感染出生24h内的乳鼠建立模型;以RT-PCR法测定血浆病毒载量;流式细胞术测定CD4+和CD8+细胞的百分数,计算CD4+/CD8+细胞的比值;镜检计数外周血白细胞、红细胞和骨髓有核细胞数;测定脾脏、胸腺的重量并计算脏体系数。结果:与正常对照组相比,模型组小鼠血浆病毒载量较高,CD4+细胞百分率、红细胞和骨髓有核细胞数以及胸腺系数显著降低,而白细胞数和脾脏系数显著升高;与模型组相比,免疫力素在1和10g·kg-1剂量下血浆病毒载量显著降低,条带亮度较暗;免疫力素0·1,1,10g·kg-1剂量组均显著升高模型动物CD4+细胞的百分率、CD4+/CD8+比值、红细胞、骨髓有核细胞数以及胸腺系数,降低脾脏系数。结论:免疫力素不仅具有一定的抗病毒作用,而且对免疫功能有一定的调节作用,提示其对艾滋病可能有较好的治疗作用。Objective： To investigate the antivirus effects of Mian-Yi-Li-Su （MYLS）, a compound Chinese Medicine, on L6565 murine leukemin virus and discuss the possibility of application of MYLS on AIDS therapy. Methods： The model was set up by infecting infant mice with L6565 murine leukemin virus. Quantitative RT-PCR method was used to measure viral load in plasma. The percentage of CD4 ^＋and CD8^＋ lymphacytes was estimated by flowcytometric method and the ratio of CD4^＋/CD8^＋ was calculated. The number of WBC, RBC and marrow karyote was counted. Indices of immune organs were weighed and calculated. Results： Compared with control, the viral load in plasma in the mice infected with the virus was at high level. The percentage of CD4^＋ lymphocyte, the number of RBC and marrow karyote, and thymus indices were significantly decreased in the model mice, while the number of WBC and spleen indices were remarkable increased. However, at 1 and 10 mg. kg^-1 , compared with model group, the viral load in plasma was significantly decreased by MYLS. And at 0.1 - 10 mg.kg ^-1 , the percentage of CD4^＋ lymphocyte, the ratio of CD4^＋/CD8^＋ , thymus index, the number of RBC and marrow karyote were remarkably increased, some of the items even recovered to normal level, but spleen indices were remarkably decreased by MYLS. Conclusion： These results suggest that MYLS has not only antivirus effects, but also immunomodulative effect. MYLS has a potential therapeutic effect on AIDS.
