摘  要:目的探讨银杏叶提取物(EGb761)对视网膜光损伤后感光细胞的保护作用。方法72只Sprague-Dawley(SD)大鼠随机分为正常对照组、模型组、模型+生理盐水组和模型+EGb761组,每组各18只大鼠。模型组、模型+生理盐水组和模型+EGb761组暗适应24h后持续光照6h,光照强度为(2740±120)lx,建立光损伤模型。模型+EGb761组和模型+生理盐水组分别于光照前7d每日腹腔注射0.35%EGb761(100mg/kg)和相应体积的生理盐水,光照后继续给药14d;于光照后4d对各组进行视网膜原位凋亡细胞检测,并于光照后7、14d作组织病理学检查并计数视盘上、下方视网膜外核层(ONL)感光细胞层数。结果光照后4d,除正常对照组外,其余各组ONL均出现凋亡感光细胞,但模型+EGb761组ONL凋亡感光细胞数明显少于模型组和模型+生理盐水组。光照后7d,模型组和模型+生理盐水组感光细胞核层数均为3~4层,模型+EGb761组为7~8层;模型+EGb761组平均感光细胞核层数(6.92±0.82)少于正常对照组(8.40±0.95)(t=-1.416,P〈0.05),但显著多于模型组(5.96±1.36)和模型+生理盐水组(5.90±1.40)(t=1.024,1.084;P〈0.05)。光照后14d,模型组和模型+生理盐水组感光细胞核为0~1层,而模型+EGb761组仍存有3~4层;模型+EGb761组平均感光细胞核层数(5.52±1.06)仍显著多于模型组(3.44±2.15)和模型+生理盐水组(3.37±1.91)(t=2.082,2.146;P〈0.05)。结论EGb761能部分抑制视网膜光损伤后感光细胞凋亡,对感光细胞具有一定的保护作用。Objective To observe the protective effect of ginkgo biloba extract (EGb 761), a free radical scavenger, on the photoreceptor cells after light-induced retinal damage. Methods Seventy-two female Sprague-Dawley (SD) rats were randomly divided into 4 groups: normal control group, lightinduced retinal damage model group, model + physiological saline group, and model + EGb 761 group, with 18 rats in each group. All of the rats except the ones in the control group were exposed to white light at (2740±120) lx for 6 hours after the dark adaptation for 24 hours to set up the light-induced retinal damage model. Rats in model + physiological saline group and model + EGb 761 group were intraperitoneally injected daily with physiological saline and 0.35% EGb 761 (100 mg/kg), respectively 7 days before and 14 days after the light exposure. Apoptosis of photoreceptor cells was detected 4 days after light exposure; 7 and 14 days after light exposure, histopathological examination was performed and the layer number of outer nuclear layers (ONL) on the superior and inferior retina was counted. Results Four days after light exposure, the apoptosis of photoreceptor cells was found on ONL in model, model + physiological saline and model+EGb 761 group, and was obviously less in model + EGb 761 group than in model and model+physiological saline group. Seven days after light exposure, the layers of ONL on the superior retina were 3 to 4 in model and model+physiological saline group, and 7 to 8 in model+ EGb 761 group; the mean of the layer number of ONL in model+EGb 761 group (6.92±0.82) was less than that in normal control group (8.40±0.95) (t =-1. 416, P〈0.05), but significantly more than that in model (5. 96±4 1. 36) and model+physiological saline group (5. 90± 1.40)(t = 1. 024, 1. 084; P〈0.05).
