摘 要:采用聚丙烯酰胺凝胶垂直板电泳法对产自黑龙江水系的黄颡鱼同功酶进行了分析.结果表明:在黄颡鱼的性腺、眼、肌肉、肝、心、肾6种组织中,10种酶(ADH,EST,GDH,IDH,MDH,LDH,POD,-αAMY,G-6-PDH和SOD)的同功酶谱均存在明显的组织特异性.其中的5种酶在雌雄性别间也存在着显著的差异.这种雌雄性别间存在的较为明显、稳定的酶谱差异可以作为黄颡鱼性别鉴定的一项参考指标.10种酶共记录出25个基因位点,其中Adh-1,Adh-3,Gdh-1,Gdh-2,Pod-2和Est-1为多态位点.黄颡鱼群体的多态位点百分数为24.00%(P0.99),平均预期杂合度和平均实际杂合度分别为0.096 2和0.005 0,极显著地偏离了Hardy-Weinberg平衡;平均有效等位基因数为1.24,在目前硬骨鱼类多样性研究资料中处于下限值;多态位点的固定指数为0.983 5,杂合子处于较深程度的缺失状态.从以上数据可以看出,这批引自黑龙江水系的黄颡鱼群体的种质较为纯化. Vertical polyacrylamide gel electrophoresis was used to investigate the tissue-specific and genetic structure of Pelteobagrus fulvidraco in Heilongjiang system.In the research,ten isozymes(ADH,EST,GDH,IDH,ME,MDH,LDH,POD,α-AMY,G-6-PDH,and SOD) in six kinds of tissues(gonad,eye,muscle,liver,heart and kidney) were investigated.The results showed that the expression of all the isozymes studied was highly tissue-specific in Pelteobagrus fulvidraco.The electrophoretic patterns of isozymes showed sex-related difference between male and female Pelteobagrus fulvidraco were apparently in 5 isozymes expression.It can be used as a standard of identity the male and female Pelteobagrus fulvidraco.Then in the genetic analysis of all the isozymes in muscle and liver of Pelteobagrus fulvidraco,twenty five enzymecoding loci were examined,of which six loci were polymorphic,i.e.,Adh-1,Adh-3,Gdh-1,Gdh-2,Pod-2 and Est-1.The proportion of polymorphic loci was 24.00%.The average values of expected and observed heterozygosity were 0.096 2 and 0.005,which strayed from Hardy-Weinberg equilibrium apparently.The average effective number of alleles was 1.24,which was at lower limit of the teleostean.Genetic deviation index was 0.983 5,which showed heterozygotes absent seriously.The data invoved indicated that the Pelteobagrus fulvidraco population which from Heilongjiang system was rather pure.
