以地高辛标记探针研究人16型乳头瘤病毒感染与良性前列腺增生症的相关性

摘　　要：目的用分子生物学方法制备地高辛标记１６型人乳头瘤病毒（ＨＰＶ１６）ＤＮＡ探针，对ＨＰＶ与良性前列腺增生症（ＢＰＨ）的相关性进行探讨。方法将质粒ｐＨＰＶ１６经ＢａｍＨＩ酶切，Ｖ－形槽电洗脱回收ＨＰＶ１６ＤＮＡ，随机引物法进行标记。用斑点杂交对探针的质量参数进行评估，并对２９例ＢＰＨ和７例正常前列腺组织进行了ＨＰＶ１６ＤＮＡ检测。结果表明本探针灵敏特异、经济实用。ＢＰＨ组和正常对照组的阳性率分别为６２．１％和１４．３％，两者差异有显著性（Ｐ＜０．０５）。结论ＢＰＨ可能是ＨＰＶ感染的又一男性宿主，ＨＰＶ１６感染可能在ＢＰＨ中起着一定的病因学作用。Objective\ The etiology and molecular mechanism of benign prostatic hyperplasia(BPH) are unclear.In the past few years,human papillomavirus(HPV)has been linked with urogenital carcinomas.In this study preparation of Dig HPV 16 DNA probe by molecular methods and elucidation of the relationship between HPV 16 and BPH were carried out..\ \ Methods\ The pHPV 16 recombinant plasmid isolated from E.coli HB 101 by improved alkaline lysis method was cut by BamHI,HPV 16 DNA recovered by electroeluting with V shaped groove was labelled by random priming with digoxigenin dUTP.Dot blot was performed for evaluating the specificity and sensitivity of the probe.Twenty nine cases with BPH and 7 cases with normal prostate tissue were detected for HPV 16 DNA with Dig HPV 16 DNA probe.\ \ Results\ The results suggest that Dig HPV 16 DNA probe is sensitive,specific,economic and practical.The positive rate in the patients with BPH(62 1%)was significantly higher than that in the normal group(14 3%)( P <0 05).\ \ Conclusions\ BPH tissue may be a reservoir for HPV sexual transmission and HPV 16 infection may have an important role in the etiology of BPH.
