摘 要:利用静电吸附作用,把乙型肝炎表面抗体(HBsAb)固定在邻氨基苯甲酸/亚甲基蓝及纳米金修饰的玻碳电极表面,从而制得高灵敏的、用于检测乙肝表面抗原(HBsAg)的电流型免疫传感器.通过循环伏安法考察了电极的电化学特性并对电极的性能进行了详细的研究.用循环伏安法测得HBsAg的线性范围为3.O~250ng/mL,检出限为1.0ng/mL.该法制备的免疫传感器具有制备简单、灵敏度高、重现性和选择性好等特点.A highly sensitive amperometric imrnunosensor has been developed for the detection of HBsAg (hepatitis B surface antigen) based on fixation of HBsAb (hepatitis B surface antibody) onto the surface of a glassy carbon electrode mediated by anthranilic acid, gold nanoparticles and methylene blue through electrostatic adsorption. Cyclic voltammetry was used to study the electrochemical properties of the electrode and showed that the immunosensor was highly sensitive to HBsAg, with a detection limit of 1.0 ng/mL and a linear range of 3.0 to 250 ng/mL. This method has good sensitivity, reproductivity and selectivity and could be easily used in HBsAg detection.
