摘  要:目的探讨甲氧沙林治疗白癜风的可能机制。方法正常人黑素细胞来自包皮环切术切取的包皮,用包被纤维黏连蛋白的48孔培养板检测黑素细胞粘附,用带微孔膜的Transwell培养板研究黑素细胞迁移,用RT-PCR方法检测基质金属蛋白酶-2(MMP-2)mRNA表达。结果50 nmol/L和100 nmol/L甲氧沙林可促进黑素细胞粘附(P<0.05)和迁移(P<0.05),100 nmol/L对黑素细胞促迁移作用最强(P<0.01),而且10 nmol/L甲氧沙林即可明显促进MMP-2 mRNA表达(P<0.01)。在一定浓度下,甲氧沙林促黑素细胞粘附、迁移和MMP-2 mRNA表达呈剂量依赖性,在不同浓度组间比较差异均有统计学意义(P<0.01、P<0.05和P<0.01)。结论甲氧沙林通过促进MMP-2的表达来促进黑素细胞粘附和迁移。Objective To investigate the possible mechanism of 8-methoxypsoralen in the treatment of vitiligo. Methods Human melanocytes were obtained from normal human foreskins, and 48-well culture dishes coated with fibronectin were used to perform melanocyte adhesion assay. Motility was assessed using the Transwell micropore filter method. RT-PCR was employed to measure MMP-2 expression. Results The 8-methoxypsoralen at 50 and 100 nmol/L could significantly promote melanocyte adhesion and migration ( P 〈 0.05 ), with the best work concentration at 100 nmol/L. The expression of MMP-2 mRNA was significantly increased by 8-methoxypsoralen at 10 nmol/L only ( P 〈 0.05 ). Within a certain range of concentration, 8-methoxypsoralen exerted its functions in a dose dependent manner ( P 〈 0.01 for adhesion, P 〈 0.05 for migration, P 〈 0.01 for MMP-2 expression ). Conclusions The efficacy of 8-methoxypsoralen in the treatment of vitiligo may be partially due to its promoting effects on melanocyte adhesion and migration, and the effect on migration may be carried out partly by enhancing the expression of MMP-2.
