摘　　要：目的明确肠嗜铬样细胞(ECL细胞)上前列腺素E(PGE)受体的表达类型和定位分布。方法分离纯化大鼠ECL细胞,观察米索前列醇、恩前列素及4种PGE受体亚型(EP1、EP2、EP3、EP4)特异激动剂(分别为ONO-DI-004、ONO-AE_1-259、ONO-AE-248、ONO-AE_1-329)对ECL细胞胃泌素诱导的组胺分泌的影响。采用逆转录-聚合酶链反应(RT-PCR)和荧光免疫细胞化学技术检测ECL细胞上4种PGE受体亚型。结果米索前列醇、恩前列素以及ONO-AE-248(1×10^(-10)～1×10^(-7)mol/L)均能明显抑制ECL细胞胃泌素诱导的组胺分泌(P值均<0.01),而ONO-DI-004、ONO-AE_1-259、ONO-AE_1-329在相同浓度下对胃泌素诱导的组胺分泌无明显抑制作用(P值均>0.05)。RT-PCR和荧光免疫细胞化学技术证明ECL细胞上同时有EP1和EP3的表达,前者的分布以胞核为主,而后者则主要分布于胞膜和胞质。结论ECL细胞同时表达EP1和EP3,两者的细胞内定位不同。EP3介导PGE对ECL细胞胃泌素诱导的组胺分泌的抑制作用。Objective To determine the subtype and distribution of prostaglandin E receptor（PGE） in enterochromaffin-like cells （ECL cells）. Methods The ECL cells were isolated and purified. The effects of misoprostol, enprostil and the specific agonists （ONO-DI-004, ONO-AE1-259, ONO -AE-248, and ONO -AE1- 329） of 4 subtypes of PGE receptors（EP1, EP2, EP3 and EP4） on the gastrin-induced histamine release from ECL cells were studied. The 4 subtypes of PGE receptors in ECL cells were examined by RT-PCR and fluorescent immunocytochemistry techniques. Results Misoprostol, enprostil, and the specific EP3 agonist ONO-AE- 248 （1 × 10^-10mol/L - 1 × 10^-7 mol/L） significantly inhibited the gastrin-induced histamine secretion by ECL cells; the same concentrations of specific agonists of EP1, EP2, or EP4 had no inhibitory effects on the gastrininduced histamine secretion. RT-PCR and fluorescent immunocytochemistry results showed that both EP1 and EP2 were present in ECL cells, with the former mainly found in the nuclei and the latter in the membrane and cytoplasm. Conclusion Both EP1 and EP3 are present in ECL cells, but with different locations. EP3 mediates the inhibitory effects of prostaglandin E on the gastrin-induced histamine secretion from ECL cells.
