摘  要:目的了解胚胎低氧环境下动脉导管平滑肌细胞(ductus arterious smooth muscle cell,DASMC)、肺小动脉平滑肌细胞(small pulmonary artery smooth musclecell,SPASMC)及相对照的主动脉平滑肌细胞(aorta smooth muscle cell,AOSMC)内钙离子浓度的变化差异。方法培养孕期120d左右胎羊的DASMC、SPASMC和AOSMC。应用特异性的Ca2+荧光指示剂Fluo-3-AM负载细胞,通过激光共聚焦显微镜动态观察模拟胚胎低氧环境下三类细胞游离Ca2+浓度的变化。结果DASMC从正常氧环境转换到低氧环境后3min,细胞内荧光强度(FI)开始降低(P<0.01),并在第8min降至最低点;SPASMC从正常氧环境转换到低氧环境后2min,FI开始升高,并在第9min升至最高点;AOSMC从正常氧环境转换到低氧环境后,其FI浓度保持不变(P>0.05)。结论在胚胎低氧环境下,DASMC胞内游离钙离子浓度降低、SPASMC胞内游离钙离子浓度升高,而AOSMC胞内游离钙离子浓度保持不变。Objectives To determine the changes of intracellular calcium concentration ([Ca^2+] i) in fetal lamb ductus arteriosus smooth muscle cells (DASMC) , small pulmonary artery smooth muscle cells (SPASMC) and arota smooth muscle cells (AOSMC) under hypoxic conditions. Methods DASMC, SPASMC and AOSMC of fetal lamb at 120-day-old were cultured. Cells were pre-loaded with fluo 3-AM and laser scanning confocal microscopy was used to measure the [Ca^2+]i of these cells under hypoxic conditions. Results The intracellular fluorescence intensity (FI) of DASMC declined at 3 rain after hypoxia (P 〈 0.01) and reached its lowest level at 8 rain. The intracellular FI of SPASMC increased at 2 rain after hypoxia (P 〈 0.01 ) and reached its peak at 9 min. The intracellular FI of AOSMC did not change significandy throughout the experiment (P 〉 0.05). Conclusions Under hypoxic conditions, fetal lamb DASMC [Ca^2+]i decreases while SPASMC [Ca^2+]i increases and AOSMC [Ca^2+]i does not change significantly.
