摘  要:目的:提取分离并测定山药多糖的含量。方法:依次用水提取,乙醇和十六烷基三甲基溴胺盐沉淀法得到粗多糖;用葡聚糖凝胶柱层析和高效液相层析纯化多糖并测定分子量;采用HPLC法测定山药总多糖含量。结果:从山药中得到两个均一多糖,相对分子质量分别为62000和7300Dal;HPLC法测得山药总多糖含量为16.42%。结论:采用本实验提取方法可有效提取分离出纯化山药多糖,利用HPLC法测定山药总多糖含量结果与其他测定方法比较,结果准确,可作为山药总多糖含量测定的方法。OBJECTIVE To isolate and determine the contents of polysaccharides from dioscorea opposite. METHODS Crude polysaccharides were obtained with water extraction, alcohol and CTAB precipitation successively, gelcolumn chromatographyon SephadexG-100 and HPLC analysis were used to purify the polysaccharides and determine their molecular weights, and to determine the contents of polysaccharides. RESULTS Two homogenous polysaccharides were isolated from the root of dioscorea opposita and the molecular weights of S1 and S2 were 62 000 and 7 300 Dal respectively; The contents of polysaccha- rides were 16. 42%. CI)NCLUSlON The polysaccharides can be extracted and isolated by the methods of the experiment, compared the determine method by HPLC with other methods, the results of the contents of polysaccharides were accurate, and the methods can be used to determine the contents of polysaccharides.
