摘　　要：目的探讨5-溴脱氧尿嘧啶核苷(BrdU)标记脂肪成体干细胞(adipose-derived adult stem cells,ADASCs)的可行性。方法将兔脂肪组织用I型胶原酶消化,分离培养ADASCs,第三代ADASCs融合约50%时,加入终浓度25mol/L的BrdU孵育48h,BrdU免疫组化染色,检测标记情况。然后通过显微镜下形态学观察、台盼蓝排斥试验、MTT试验,观测BrdU对ADASCs生长增殖的影响。结果BrdU对ADASCs的生长增殖基本没有影响。结论BrdU标记ADASCs可行且较理想。Objective To study the feasibility of labeling adipose-derived adult stem cells with BrdU. Methods The adipose-derived adult stem cells were obtained from the rabbit adipose tissue that was digested with collagenase Type I. Then they cultured and expanded in control medium. The third passage was incubated with BrdU at 251amol/1 for 48h when the cells confluenced about 50%. Then immunohistochemical assay was performed to evaluate the cellsfor proper labeling. Then by the morphology under the microscope, trypanblau testing, MTT testing, we surveyed the influence of BrdU on the growth of ADASCs. Results It makes no difference in the proliferation of ADASCs whether BrdU is used to labell the cells. Conclusions BrdU is feasible and ideal to labeling adipose-derived adult stem cells.
