摘 要:[目的]探讨血管内皮细胞生长因子(VEGF)、VEGF受体(VEGFR2)及P53在门脉高压大鼠舌下异常络脉的表达及意义。[方法]CCl4注射法制备肝硬化门脉高压(CPH)大鼠模型,以正常大鼠为对照;门静脉部分结扎制备肝前型门脉高压症(PHPH)模型,以假手术大鼠为对照;不同时点观察舌下络脉形态、颜色,并测定门静脉压力;免疫组织化学En Vision方法检测大鼠舌下血管VEGF、VEGFR2及P53的表达。[结果]随着门静脉压力的升高,CPH组和PHPH组大鼠舌下血管出现明显形态学和颜色改变;免疫组化结果显示正常对照组舌下血管未见VEGF、VEGFR2及P53的表达,假手术组大鼠在术后7 d出现VEGF的微量表达,余时点均为阴性;CPH和PHPH大鼠术后不同时点可见VEGF、VEGFR2及P53蛋白表达,并持续存在。[结论]门静脉压力变化可能是异常舌下络脉形成的一个重要因素,舌下血管内皮上VEGF、VEGFR2及P53的高表达可能参与了这一过程。[Objective] To evaluate the expression of vascular endothelial growth factor (VEGF), vascular endothelial growth factor receptor-2 (VEGFR2) and P53 in sublingual collaterals of portal hypertension rats. [Methods] The models of cirrhotic portal hypertension(CPH) were established in SD rats by 50% carbon tetrachloride (CCl4) subcutaneous injection. CCl4 injected rats were compared with normal control ( NC ) rats. The models of prehepatic portal hypertension(PHPH) were established in SD rats by partial portal vein ligation. PHPH rats were compared with sham operation ( SO ) rats. Morphous and color of sublingual collaterals were observed in different time spots, and portal venous pressure were determined. Expression of VEGF, VEGFR2 and P53 in sublingual collaterals of the rats were detected with immunohistochemistry. [Results] The sublingual veins in rats of CPH group and PHPH group changed quickly in the morphology and the color, while portal venous pressure was increased. There was no VEGF,VEGFR2 and P53 expression in the sublingual collaterals of NC group. In SO group, VEGF expression was very low and appeared at the 7^th day, but at other time spots were negative. VEGF, VEGFR2 and P53 expression in CPH group and PHPH group can be detected at different days, and existed after operation. [Conclusion]The changes of portal vein pressure are an important factor in formation of abnormal sublingual collaterals. The increased expression of VEGF, VEGFR2 and P53 in sublingual collaterals may act as a promoter of the process.
