子前期胎盘组织差异表达基因PP2ACβ的初步研究

摘  要:目的寻找并克隆子前期胎盘组织高表达基因蛋白磷酸酶2A催化亚基β(PP2ACβ),研究其mR-NA及蛋白水平的表达,探讨其在子前期中的作用。方法标本取自2004年5月至2004年12月天津中心妇产科医院重度子前期及正常孕妇各30例。应用荧光mRNA差异显示技术(FDD)发现并克隆子前期胎盘组织差异表达的基因PP2ACβ,进一步应用半定量RT-PCR及免疫组化技术分析差异表达基因PP2ACβmRNA及其蛋白水平PP2A的表达。结果通过FDD发现PP2ACβ基因在子前期胎盘组织中高表达,半定量RT-PCR证实PP2ACβmRNA在子前期胎盘组织中的表达(0.888±0.104)高于正常胎盘组织(0.692±0.099),差异有统计学意义(P<0.05),免疫组化验证其蛋白水平PP2A的表达(0.14±0.02)亦高于正常(0.12±0.02),差异有统计学意义(P<0.05)。结论FDD可有效用于筛选子前期胎盘组织差异表达基因;PP2A可能通过促进胎盘滋养层细胞的凋亡及阻碍血管生成而参与子前期的发病。Objective To search and clone the highly expressed protein phosphatase 2A catalytic subunit - beta (PP2ACβ) gene in the placenta of preeclampsia, and study its expression both in mRNA and protein level, to investigate its role in the pathogenesis of preelampsia. Methods The differentially expressed genes from placenta of preeclampsia were found by fluorescent differential display ( FDD ) and cloned, PP2ACβ was one of the up - regulated genes. PP2ACβmRNA was measured by semi - quantitative RT - PCR in the placentas of 30 preeclampsia and 30 normal pregnant women, its protein was detected by immunohistochemistry (IMH), Results The expression of PP2ACβ was up - regulated in placentas of preeclampsia, PP2ACβmRNA was significantly higher in placentas of preeclampsia than that in the matched normal placentas(0. 888 ± 0. 104,0. 692 ± 0. 099;P 〈 0.05). The protein of PP2A was higer in placentas of preeclampsia than that in the matched normal placentas (0.14 ± 0. 02,0. 12 ± 0. 02;P 〈 0. 05). Conclusion The genes differentially expressed between the placentas of preeclampsia and normal pregnancy can be found by FDD effectively. PP2A may play a role in the pathogenesis of preeclampsia.
