摘  要:目的:探讨具有α糖苷酶抑制活性的蚕沙提取物在动物模型中的抗糖尿病作用。方法:实验选用正常小鼠及四氧嘧啶高糖小鼠,比较蚕沙提取物对蔗糖、淀粉及葡萄糖负荷后血糖升高的影响。用四氧嘧啶高糖大鼠,测定给药4周后蚕沙提取物对血糖、尿糖、血脂、果糖胺、血清N-乙酰-β-D-氨基葡萄糖苷NAG酶活性、晶体和坐骨神经中山梨醇等含量的影响。结果:蚕沙提取物能明显降低正常小鼠和高糖小鼠蔗糖或淀粉负荷后的血糖峰值及血糖曲线下面积(AUC),并使血糖峰值后移,但对葡萄糖耐量无影响。长期实验中,蚕沙提取物可明显改善高糖大鼠的三多症状,使空腹血糖、非禁食血糖、血清果糖胺浓度、血脂及尿糖等明显低于对照组,另外血清N-乙酰-β-D-氨基葡萄糖苷NAG酶活性、坐骨神经中山梨醇含量及红细胞中还原型谷胱甘肽(GSH)含量也有明显改善。结论:蚕沙提取物具有α-糖苷酶活性抑制作用,改善糖尿病动物的糖、脂代谢异常,可能有益于糖尿病慢性并发症的防治。Objective: To evaluate the anti-diabetic potency of the extraction from Cansha (Bombyx mori L. ), a Chinese traditional medicine, with the alpha-glucosidase inhibition activity. Methods: The effects of Cansha extraction on tolerances of sucrose, starch and glucose were compared in normal and alloxan-induced diabetic mice. Biochemical variables such as the fasting blood glucose, urine glucose, the serum lipid, serum fructosamine, serum NAG activity, sorbitol content in lens and sciatic nerve were determined in alloxan-induced diabetic rats treated with the extraction for 4 weeks. Results: Cansha extraction significantly decreased the blood glucose peak and the AUC after sucrose or starch load. It also shifted the peak to right, but there was no impact in glucose tolerance test. In the long-term experiment, Cansha extraction improved 3 P symptoms (polyuria, polidipsia and polyphagia) , and obviously reduced the fasting and post-prandial blood glucose, serum fructosamine, the serum lipid and urine glucose in hyperglycemic animals. It also significantly reduced the serum NAG activity, sorbitol content in sciatic nerve, GSH content in erythrocyte. Conclusion: Cansha extraction shows strong inhibitory activities against alpha-glucosidase in intestine. It can ameliorate the disorder of glucose and lipid metabolism and improve or delay diabetic complication.
