摘 要:目的观察消渴颗粒剂对高糖刺激后系膜细胞增殖和转化生长因子-β1(TGF-β1)表达的影响。方法培养正常大鼠系膜细胞,高浓度葡萄糖刺激系膜细胞,加入中药含药血清后,用MTT法和激光共聚焦显微镜分别观察系膜细胞增殖和TGF-β1的表达。结果消渴颗粒剂含药血清在48h可抑制高糖对大鼠系膜细胞的促增殖作用,在72、96 h可对抗高糖对系膜细胞增殖的抑制作用,96 h最为明显(P<0.05)。高糖刺激后TGF-β1在肾小球系膜细胞内表达增多,消渴颗粒剂明显减少系膜细胞内TGF-β1表达(P<0.05)。结论消渴颗粒剂可降低高糖致系膜细胞增殖,抑制高糖引起的TGF-β1过度表达,这可能是其防治糖尿病肾病的作用机制。Objective To investigate the mechanism of Xiaoke Granules for treating diabetic nephropathy by studying the influence of Xiaoke Granules on proliferation and TGF-β1 expression in mesangial cells. Methods Rat mesangial cells (MCs)were cultured in vitro, and were exposed to high glucose concentration. The effect of drug serum of Xiaoke Granules on MCs proliferation and TGF-β1 expression in MCs were identified by MTT colorimetric method and laser confocal scaning microscopy respectively. Results The drug serum of Xiaoke Granules inhibited MCs proliferation promoted by high glucose at 48 hour, and reversed the depressed proliferation induced by high glucose at 72 hour and 96 hour, with significant difference at 96 hour( P 〈 0.05). TGF-β1 expression was high in MCs stimulated by high glucose ,while drug serum of Xiaoke Granules depressed this high expression of TGF-β1 in MCs( P 〈 0. 05 ). Conclusion Xiaoke Granules can reverse the influence of high glucose on MCs proliferation, and can inhibit the high expression of TGF-β1 induced by high glucose. This may be the key function of Xiaoke Granules on treating diabetic nephropathy.
