重组痘苗病毒VMS11HAV25株的毒力研究

摘  要:对带有甲肝病毒抗原基因的重组痘苗病毒VMS11HAV25株与其亲本病毒天坛株和JingC-1系减毒株进行了毒力比较,结果表明,VMS11HAV25株对小鼠神经毒力和尾静脉注射后的全身症状以及家兔皮内反应都比天坛株有明显减弱。在40℃培育下鸡胚尿囊膜(CAM)上形成的痘疱数也比天坛株有明显减少。VMS11HAV25株在37℃培育下CAM上形成的痘疱较大,家兔皮内接种后出现有小面积的皮肤坏死性损伤,因此认为,VMS11HAV25株的残余毒力高于JingC-系,介于天坛株和JingC-1系减毒株之间。The virulence of VMS11HAV25 strain, a recombinant vaccinia virus strain harbouring hepatitis A virus gene, was compared with those of its parental strain (Tiantan strain) and attenuated Jing C-1 strain. The result showed that the intracerebral and systemic virulence of VMS11HAV25 strain in mice and the intracutaneous virulence in rabbits were significantly weaker than that of Tiantan strain. After chick embryo allantoic membranes (CAM) were infected with the 3 strains and cultured at 37℃ and 40℃, the number and size of the pocks formed on the surface of CAM were compared. The number of pocks induced by VMS11HAV25 strain at 40℃ was significantly decreased compared with that induced by Tiantan strain, however, the sizes of them at 37℃ were significantly larger than that induced by Jing C-1 strain at the same temperature. In addition, small area of necrotic lesion of skin was observed in rabbits intracutaneously injected with VMS11HAV25 strain. The authors considered that the residual virulence of VMS11HAV25 strain was stronger than that of Jing C-1 strain but weaker than that of Tiantan strain.
