摘 要:目的探讨正丁基苯酞(NBP)对血管性痴呆(VD)小鼠空间学习记忆及海马细胞内游离钙([Ca^2+)]i)浓度、钙-钙调蛋白依赖性蛋白激酶Ⅱα(CaMKⅡα)蛋白表达的影响。方法采用双侧颈总动脉线结法制备小鼠VD模型,服用NBP(120mg·kg^-1·d^-1)组为治疗组;并设假手术组作为对照。运用跳台和水迷宫装置观测各组小鼠行为学变化,流式细胞仪检测海马细胞内[Ca^2+)]i浓度变化,Western Blot印迹方法测定CaMKⅡα蛋白的表达。结果VD模型组小鼠学习记忆成绩下降,海马细胞内[Ca^2+)]i浓度增高,CaMKⅡα蛋白表达水平降低,与假手术组比较差异有统计学意义(P〈0.05);NBP治疗组小鼠学习记忆成绩改善,海马[Ca^2+)]i浓度降低,CaMKⅡα蛋白表达水平增高,与模型组比较差异有统计学意义(P〈0.05),与假手术组比较,差异无统计学意义(P〉0.05)。结论NBP可降低VD小鼠海马神经细胞内游离[Ca^2+)]i,提高CaMKⅡα的表达,改善其学习记忆能力。Objective To explore the effect of dl-3n-butylphthalide (NBP)on spatial learning and memory, the intracellular free calcium ([Ca^2+] i) concentration and the calcium/calmodulin -dependent protein kinase Ⅱα (CaMKⅡα) expression in hippocampus of mice with vascular dementia (VD)and to explore the molecular mechanisms of NBP on VD. Methods The mice received the ligation of bilateral common carotid arteries to establish the VD models. The treatment group were administrated with NBP(120 mg · kg^-1 · d ^-1 ) after the establishment of VD model. Those animals with the shamed operation were taken as control group. The changes of behavior were observed through the step-down avoidance test and water maze test. Meanwhile, the intracellular [Ca^2+]i and the expression of CaMKⅡα were determined by flow cytometry and Western blot respectively. Results The abilities of learning and memory in model group was poor, the concentration of intracellular [Ca^2+]i in hippocampus was increased, the expression of CaMKⅡα was decreased, compared with the shamed operation(all P〈0.05). The abilities of learning and memory in NBP treatment group was improved, the [Ca^2+] i were decreased, the expression of CaMKⅡα was increased, compared with model group(P〈0.05), but no significant differences was found between the NBP treatment group and shamed operation group(P〉0. 05). Conclusions NBP can decrease the concentration of intracellular [Ca^2+ ]i and increase the expression of CaMKⅡα, meanwhile, it can improve the learning and memory ability of VD mice.
