摘 要:目的:探讨草酸钙尿石症患者肾组织中维生素K依赖性羧化酶(VKDC)基因mRNA表达水平及其与草酸钙尿石形成的关系。方法:以TaqMan实时荧光定量PCR法检测19例草酸钙尿石症肾组织(尿石组)与21例非尿石肾组织(对照组)标本中VKDC基因mRNA的表达,采用2-△△Ct法比较两组标本中VKDC基因mR-NA表达的差异。结果:尿石组与对照组荧光定量检测△Ct值分别为8.996±1.667与6.023±1.234(P<0.05),尿石组VKDCmRNA表达明显降低。结论:草酸钙尿石症患者肾组织中VKDC基因表达明显下降,其表达水平的变化可能与草酸钙结石形成有关。Objective:To study the gene expression of vitamin k dependent carhoxylase (VKDC) in patients with calcium oxalate urolithiasis and evaluate the correlation between the quantity of VKDC and the formation of calcium oxalate calculus. Methods: Using Taqman real-time reverse transcription-polymerase chain reaction (RT-PCR) and 2^△△Ct, method, the expression of VKDC mRNA were evaluated in renal tissue specimens from patients with calcium oxalate urolithiasis (urolithiasis group, n= 19) and non-urolithiasis (control group, n= 21). Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) was used as the housekeeping gene. Results: The amount of VKDC mRNA in urolithiasis group was lower than in the controls, indicated by significantly higher delta threshold cycles (△Ct) in RT-PCR for the VKDC gene in the urolithiasis renal tissue, with mean ± SD of 8. 996±1. 667 and 6. 023±1. 234(P〈0. 01), respectively. VKDC mRNA was on average 7.86-fold(range, 2. 47 to 24.93-fold)underexpressed in urolithiasis group compared to controls. Conclusions:These data demonstrate that the gene expres- sion of VKDC is decreased in renal tissues of patient with calcium oxalate urolithiasis, and may he involved in the formation of calcium oxalate calculus.
