摘  要:为了提高肾移植的成功率,降低环孢素A(CsA)的肾毒性,应用中药川芎及川芎嗪进行了防止肾毒性的研究。给SD雄性大鼠灌服CsA,制成肾中毒模型,分别经腹腔注射川芎和川芎嗪,于用药前后检测大鼠的肾功能、尿酶等指标,最后进行肾脏光镜检查。结果发现,川芎预防组大鼠的肾功能、尿酶及肾脏病理性损害均明显好转,但川芎嗪未能改善CsA引起的肾功能下降及肾小球近曲小管损害。实验证实,川芎对CsA肾中毒的发生具有预防作用,其有效成分不是川芎嗪。An experimental study was carried out in male Sprague Dawley rats on the effects of chuangxiong (ligusticum wallichi franch) and tetramethypyrazine on acute nephrotoxicity caused by cyclosporin A (CsA).The rats were divided into 4 groups:group I served as controls,group II received CsA by gavage,50mg/kg daily for 7 days,group III received CsA as above and also chuangxiong intraperitoneally,group IV received CsA and tetramethypyrazine intraperitoneally.Renal function and urinary enzymes were measured both before and on the 7th day of treatment.The animals were sacrificed on the 7th day and the kidneys were studied with light microscopy.The results indicated that CsA did produce an acute impairment of the renal function and chuangxiong could prevent the acute nephrotoxicity of CsA,both functionaly and morphologically but tetramethypyrazine could not.
