摘 要:目的建立沙棘的简单重复序列区间聚合酶链反应(ISSR-PCR)优化反应体系。方法对影响ISSR-PCR反应的各因子,如模板DNA用量、TaqDNA聚合酶浓度、镁离子浓度、dNTP浓度等进行单因素梯度实验。结果筛选出了优化的反应条件。结论建立的反应体系多态性高,系统稳定,可应用于沙棘属植物遗传多样性研究和亲缘关系分析。OBJECTIVE To establish the optimized inter- simple sequence repeat PCR( ISSR - PCR) reaction system of Hippophae rhamnoides L. METHODS The influential factors of ISSR - PCR amplification, such as template DNA dosage,Taq DNA polymerase Mg^ 2+ concentration,Mg concentration,and dNTP concentration were analyzed based on single factor gradient tests when they were applied to Hippophae L. RESULTS Each factor was optimized and determined. CONCLUSION The established system proves applicable with high polymorphism and good reproducibility, which could serve the study on genetic diversity and the analysis about gentic relationships of Hippophae rhamnoides L.
