摘  要:目的观察白藜芦醇(Res)对原代培养大鼠脑皮质神经元氧糖缺失损伤的保护作用。方法将新生大鼠脑皮质神经元原代培养至第10天,以低糖结合物理性缺氧诱导缺氧缺糖(OGD)模型,并于造模2 h后复氧复糖(RP),尼莫地平(Nim 1.0μmo.lL-1)为阳性对照,观察Res对OGD和OGD/RP的噻唑蓝(MTT)液比色吸光度和乳酸脱氢酶(LDH)释放百分率的影响,在光镜和电镜下观察神经元的形态变化。结果Res的增加对神经元OGD 1、2、4 h和OGD 2 h/RP 3、12、24 hMTT液的吸光度有显著性差异,可减少其LDH的释放,减轻神经元形态的损伤。结论Res对原代培养大鼠脑皮质神经元氧糖缺失损伤具有保护作用。OBJECTIVE To investigate the protective effects of resveratrol (Res) on injury induced by oxygen - glucose deprivation (OGD) in the primary cultured neurons of neonatal rats. METHODS In the 10th day since cerebral cortical neurons of neonatal rats were cultured, OGD model was induced by low carbohydrates plus physical hypoxia, and reperfused with oxygen - glucose deprivation (RP) 2 h later. In the experiment, Nimodipine (Nim, 1.0 μmol·L^-1) was used as positive control, MTr reduction and the release rate of lactate dehydrogenase (LDH) were measured at the different time of OGD 1, 2, 4 h, OGD 2 h/RP 3, 12, 24 h. Morphologic change of neurons were observed by light microscope and electron microscope. And the effects of Res(0. 1,1.0,10. 0 μmol·L^-1 ) on injury induced by OGD and OGD/RP were determined. RESULTS MTr reduction and the release of LDH were ameliorated by pretreatments with Res at OGD 1,2, 4 h, OGD 2 h/RP 3, 12, 24 h, and pretreatments with Res ameliorated the morphologic changes of neurons induced by OGD and OGD/RP. CONCLUSION Res may protect against injury induced by OGD in the primary cultured neurons of neonatal rats
