摘 要:肝胆宁(Oxaphenamidi,Oxa)对Fenton反应生成的羟自由基(·OH)有较强的消除作用(SC50为73.1μmol/L),其作用超过·OH的特异性清除剂甘露醇(SC50为21.3mmol/L),能够清除NBT/PMS/NADH体系产生的超氧阴离子(O2-),其SC50约为145.0μmol/L,相当于10.3U的超氧化物歧化酶(SOD,O2-的特异性清除剂);肝胆宁(150μmol/L)能有效地抑制由AAPH诱发的红细胞氧化性溶血(抑制2h以上),显示了较强的防御脂质过氧化的能力.提示肝胆宁对急慢性肝炎、胆囊炎、胆道炎等炎症、促进胆汁分泌及胆结石等的治疗作用可能与其清除自由基及防御脂质过氧化的能力有关.The effect of Oxaphenamidi (OXA) on oxygen free radicals and on oxidativehemolysis in mouse erythrocytes were studied. OXA wa, found to significantly scavenge the hydroxyl radical (-OH) produced by Fenton reaction (SG50 = 73.1umol/L),and the scavenging effectof OXA was obviously stronger than that of mannitol, a specific -OH scavenger (SC50=21.3lnm0l/L). OXA was also able to scavenge superoxide anion (O)produced by NBT/PMS/NADHsystem (SC50 = 145 .0 umol/L, which is as strong as 10.3u of SOD, a specific O scavenger). Atthe same time, OXA was also shown to have strong inhibitory effect on erythrocytes oxidativehemolysis (The hemolysis was inhibited for more than 2hrs). The results suggested that the anti-inflammation actiyity of OXA, including antihepatitis, anticholecystitis and anticholangitis,andits effeots of promoting choleresis and curing cholelithiasis may be, at least partially, related toits scavenging effects on oxygoil free radicals and the inhibitory effect on lipid peroxidation.
