摘 要:目的:以大鼠组织匀浆及肝亚细胞为材料研究海洋新结构化合物软珊瑚胆甾烯(24-亚甲基-3β、7β、19-三羟基-5-胆甾烯)的抗氧化作用。方法:组织匀浆或肝亚细胞悬液与软珊瑚胆甾烯共浴后,以VitC+FeSO4或邻苯三酚激发脂质过氧化,以丙二醛产生,还原性谷胱甘肽耗竭,膜唾液酸耗竭、线粒体膨胀作为脂质过氧化程度指标。结果:7.6、30.6、122.2μmol·L-1软珊瑚胆甾烯可抑制VitC+FeSO4激发的丙二醛产生,阻遏肝组织匀浆还原性谷胱甘肽耗竭,线粒体膨胀,但对邻苯三酚致质膜唾液酸耗竭无显著影响。结论:软珊瑚胆甾烯具有一定的抗氧化作用,但可能并无直接清除超氧阴离子自由基(O-·2)的作用。AIM: To study antioxidation of 24 methylene 3β、7β、19 trihydroxyl 5 cholestene (MTC) in tissues′ homogenates and in liver subcellulae. METHODS:Samples of tissues′ homogenates or liver subcellular suspension were pretreated with MTC, then, addition of Vit C+FeSO 4 or pyrogallol initiated lipid peroxidation. The generation of malondialdehyde, exhaustion of glutaghione in liver homogenate and sialic acid in membrane suspension, and swelling of mitochondria were examined. RESULTS:MTC could inhibit Vit C+FeSO 4 initiated lipid peroxidation in homogenates of different tissues (cerebrane, heart, lung, liver and kindey)and in liver subcellular suspensions (plasma membrane, mitochondrion, microsome) of rat. MTC had potency to prevent exhaustion of glutathione in liver homogenate and swelling of mitochondria which followed lipid peroxidation. MTC had no effect on decrease of sialic acid in plasma membrane which evoked by pyrogallol. CONCLUSION:MTC had preventive action aginst lipid peroxidation, but should have no action of scavening super oxygen anion radicals.
