摘  要:目的检测皮肤切创愈合过程中caspase-8的表达,探讨用caspase-8表达的规律推测皮肤损伤时间。方法建立小鼠皮肤切创模型,应用免疫组织化学技术和Western blot方法检测切创后不同时间皮肤中caspase-8的表达,以未切创小鼠皮肤组织作对照。结果伤后12~24h的损伤皮肤组织中部分单核细胞表达caspase-8。随伤后时间延长,caspase-8阳性细胞以单核细胞及成纤维细胞为主。结论伤后3-6h,caspase-8阳性细胞比率较低,12h后逐渐升高,3d达到高峰,以后逐渐下降。实验组中,1、3、5、7d组可见明显的caspase-8活化片段,位于分子量Marker44/42处。结论小鼠皮肤切创愈合过程中,caspase-8在损伤区单核细胞及成纤维细胞中表达,其时序性变化规律可望用于皮肤损伤时间的推断。Objective To investigate the expression of caspase-8 during the skin incised wound healing and the applicability of caspase-8 to estimation of skin wound age. Methods After model of the skin incised wound was established in mice, the expression of caspase-8 was detected by immunohistochemical and Western blotting methods. The uninjured mice were used as control. Results In the wound specimens aged from 12h to 24h post-injury, caspase-8 was identified in part of mononuclear cells (MNCs). Afterwards, the MNCs and fibroblastic cells (FBCs) accounted for the most part of the caspase-8-positive cells. Morphometrically, the ratios of the number of caspase-8 positive cells to total number of the cells in the wounds were evaluated and calculated. The ratios of the caspase-8-positive cells were low in the wound specimens aged from Oh to 3h and maximized at 3 days. Thereafter, the ratios decreased. The obvious bands of 44/42 kD were noticed at l d, 3d, 5d and 7d post-injury, indicating that caspase-8 was activated. Conclusion The results suggest that caspase-8 is expressed in MNCs and FBCs during the skin incised wound healing in mice. Caspase-8 may be used as a marker for estimating skin wound age.
