助孕胶囊对多囊卵巢大鼠卵巢细胞凋亡的影响

摘　　要：目的观察助孕胶囊对多囊卵巢(PCO)大鼠血性激素水平及卵巢细胞凋亡的影响,探讨其对PCO模型大鼠的治疗作用并分析其可能的作用机制。方法采用孕激素联合人绒毛膜促性腺激素(HCG)造模法制备PCO模型大鼠。随机分为对照组、PCO模型组、中药组(助孕胶囊)、西药组(克罗米酚),运用放免法测定血性激素水平的变化;采用TUNEL法检测卵巢组织凋亡细胞及免疫组化染色检测卵巢组织颗粒细胞Bcl-2、Bax的表达水平。结果助孕胶囊可明显降低PCO大鼠血清睾酮T、促黄体生成素LH水平,显著降低卵巢颗粒细胞凋亡率并下调卵巢组织颗粒细胞Bax蛋白的表达。结论助孕胶囊能够通过调整内分泌及抑制卵巢局部细胞凋亡,在中枢及卵巢局部协调发挥作用,从而调整PCO性腺功能,促进卵泡的发育成熟及排卵。Objective To observe the effect of Zhuyun Jiaonang （ZYJN） on procreate endocrine hormone and the expression of ovarian local cytokines in rats with polycystic ovary （PCO）, and analyze the possible therapeutic mechanism. Methods The PCO rat models were established by pregnancy hormone combined with human chorionic gonadotrophin （HCG）. Then they were randomly divided into four groups, control group, model group, Chinese crude drug group （ZYJN） and western medicine group （CC）. The levels of serum gonadal hormone were detected by radioimmunoassay. Using the immunohistochemical staining technique we detected the expression level of apoptosis regulatory protein Bcl-2, Bax and apoptotic index in ovarian granular cell with TUNEL marking methods. Results ZYJN could decrease the level of testosterone （T） and luteotropic hormone （LH） in rat serum and the expression of protein Bax. The apoptosis index （AI） of ovarian granular cells in model group was obviously higher than that in the control group and treatment group （P〈0.05）. In comparison with model group, the difference was apparent in ZYJN, whose apoptosis index （AI） of ovarian granular cells was obviously lower than the model group （P〈0.05）. Conclusion ZYJN could play a coherent role in the center and ovary by regulating the endocrine function and affecting the apoptosis of ovarian granular cell in rat ovary, thus adjusting gonadal gland and promoting the maturity and ovulation of follicle.
