复方水蛭素提取物抗移植性肿瘤的实验研究

摘　　要：目的探讨复方水蛭素提取物(CEH)对小鼠移植性肿瘤和Th1/Th2类细胞因子的作用机制,为临床用药提供理论依据。方法①CEH体内抑瘤实验:取小鼠40只,随机分组,均接种S-180瘤细胞。阴性对照组给予生理盐水,阳性对照组给予氟脲嘧啶,实验组分别给予大小剂量的CEH,连续用药14d,比较各组肿瘤生长情况,并计算抑瘤率;②检测Th1/Th2类细胞因子:取荷瘤鼠组织标本,提取细胞总RNA,紫外分光光度计测出A260及A280值;采用逆转录(RT)反应、聚合酶链反应(PCR)、电泳鉴定,及定量分析,将凝胶电泳图像输入美国Kodak凝胶分析系统,应用1D Image Analysis Software进行表达强度分析。结果CEH对小鼠移植性肿瘤具有明显的抑制作用,抑制率分别为49．06%和35．53%,其中大剂量组优于小剂量组。荷瘤鼠用药后,IL-2、IFN-λ细胞因子升高,IL-10、IL-4细胞因子降低。结论CEH对小鼠移植性肿瘤具有显著的影响,同时使荷瘤鼠低下的Th1类细胞因子增多,免疫能力提高,促使漂移失调的Th1/Th2类细胞达到平衡状态。Objective To discuss the mechanism of CEH on mouse＇s trsnsplantable tumor and CK- Thl/Th2,so as to provide academic foundation in in clinic al application.Methods （1）The experiment of restrain tumor in body of CEH：40 mouses were randomed into negative comparison group and electropositive comparison group,all of mouses were inoculated S-180 tumor cell.Saline,Cyclophosphamide and 5-FU were used respectively in negative and electropositive comparison group.The CEH which was divided into big dose and small dose was given to experimental treatment group;14 days later, the tumor＇s growth of different group were compared and the rate of restraining tumor were calculated.（2）the CK-Thl/Th2 was detected： extract cell＇s total RNA from bearing- cancer mouse＇s tissue sample; A260 and A280 were measured by uhraspectrophotometer; using RT, PCR,electrophpresis identity, authors took analysis by mount,put the GE picture into American Kodak Gel analytical system,and used ID Image Analysis Software to express the intensity analysis.Results CEH can effectively restrain the mouse＇s transplantable tumor, the rate of restraint was separatelly 49.06% and 35.3%,the group which was given big dose was better than the group which was given small dose.After the bearingcancer mouse took medicine ,CK-IL-2 and CK-IFN-γ increased and CK-IL-10 and CK-IL-4 reduced.Conclusion CEH has an obvious effect on mouse＇s transplantable tumor, at the same time,it makes the CK-Thl which was low in bearingcancer mouse＇s body to be increased,the immunity to be improved and the imbalancable CK-Thl/Th2 reach the state of balance.
