摘 要:目的观察大鼠脑出血后神经缺损症状和海马线粒体膜电位的改变及不同中医治法的干预作用。方法用Ⅶ型胶原酶于脑内定位注射诱导大鼠脑出血模型。采用神经缺损积分评定神经功能;用荧光分光光度计测定线粒体悬液的荧光强度反映线粒体膜电位。结果1.大鼠脑出血后迅速出现神经缺损症状,模型组和各中药干预组大鼠神经缺损积分于造模后4h达高峰,随后逐渐下降,至第7d仍高于假手术组和正常组(P<0.01),采用不同中医治法治疗后于第1、3、7d其神经功能缺损积分均低于模型组(P<0.05),于3、7d平肝熄风汤组积分又比风、火、痰、瘀分治各组明显降低(P<0.05)。2.脑出血大鼠线粒体膜电位于术后4h显著下降,1d时膜电位下降达高峰,3、7d逐步恢复,仍低于正常组和假手术组(P<0.01)。采取不同中医治法后,平肝熄风汤组在脑出血后,3、7d两个时间点线粒体膜电位明显高于其它治疗组和模型组(P均<0.05)。结论大鼠脑出血后线粒体膜电位下降;采用不同中医治法,均能抑制膜电位下降,改善神经功能缺损症状,但以平肝熄风汤风火痰瘀同治效果最佳。Objective To study the effects of various TOMs treatment on the changes of mitochondrial membrane potential (MMP) after intracerebral hemorrhagic (IOH) injury in rat models, and to explore the therapeutic mechanism of PGXF decoction. Methods Ⅶ type collagenase was stereotaxically injected into the globas pallidus of rats to induce intracerebral hemorrhage and spectrofluorometer was used to detect and monitor the fluorescence value changes occurred in the mitochondrial suspension, and the neurobehavioral scores were used to evaluate the neurological defect. Results l. Neurological defects were observed after IOH in rat models and the scores of neurological defects in model group received various TOMs intervention reached to peak values 4 h after onset of hemorrhage, and then decreased gradually, but were still higher than those in sham operation group and normal group on the 7th day (P〈 0.01). The equation scores of the treatment groups were lower than the model group on the 1st, 3rd and 7th days. The scores of Pinggan Xifeng decoction group were lower than the model group and other TOMs groups on the 3rd and 7th days (P〈 0.05). 2. The MMP of ICH model groups reduced significantly 4 h after onset of hemorrhage, peaked at the 1st day and then still reduced as compared with the sham - operation group and normal group during the 7th day(P〈0:01). The MMP in the PGXFD group was significantly higher than the model group and other treatment groups on the 3rd and 7th days (P〈 0.05). Conclusions MMP decreased in rats experienced with experimental IOH. Various TOM therapies can depress the decrement of MMP and improve the neurological defects, particularly Pinggan Xifeng decoction shows the best benefit.
