摘 要:目的观察异丙酚对大鼠脑复苏后海马神经元胞内游离钙[Ca2+]i水平的影响。方法26只SD大鼠随机分为三组,异丙酚组(P组、n=10),乳剂组(L组、n=10)和生理盐水组(N组、n=6)。建立窒息性心跳骤停大鼠心肺脑复苏的模型,心肺复苏成功后P组大鼠给予异丙酚10mg/kg静脉推注,继之以30mg/(kg.h)速率持续输注1h;L组大鼠则以等容积的1%脂肪乳剂按同样方式处置;N组大鼠除不作窒息、心肺复苏以外,其它操作同上两组,用等容积的生理盐水处置。复苏后2h在戊巴比妥麻醉下开颅,分离右侧海马测定神经元游离钙。结果各组大鼠之间在心肺复苏前后各时间点相应的温度、血压、呼气末二氧化碳浓度等指标的差异无统计学意义(P>0.05);和N组相比,P组、L组[Ca2+]i明显升高(P<0.05),但P组、L组之间[Ca2+]i差异无统计学意义(P>0.05)。结论心肺复苏后2h大鼠海马神经元胞内游离钙水平显著升高;异丙酚不能减缓胞内游离钙的升高。Objective To investigate the effects of propofol on concentration of intracellular free-calcium in neurons after brain resuscitation in rats.Methods 26 Sprague-Dawley rats were randomly divided into 3 groups: propofol group (P,n=10),emulsion group(L,n=10) and normal sodium group(N,n=6). The rats model of cardiopulmonary-cerebral resuscitation was established by asphyxia-induced cardiac arrest.Propofol(10mg/kg)was intraveinously injected in P group after cardiopulmonary resuscitation,and then changed infusion rate to 30mg/kg/h for one hour.Equal volume of lipid-emulsion and normal sodium was administrated in group L and group N,respectively.The rats in group N was not established the model of cardiopulmonary-cerebral resuscitation.Tight hippocampus in rats was separated undergoing anesthesia with pentobarbitone 2 hours after cardiopulmonarycerebral resuscitation,and free-calcium in neurons was determined.Results There were no statistical significance in those indexes such as temperature,blood pressure,end-tidal carbon dioxide at different time points among three groups(P〉0.05).The intracellular free-calcium in neurons in group P and group L was elevated significantly than that in group N(P〈0.05).But there were no statistical significance between group P and group L(P〉0.05).Conclusions The concentration of intracellular free-calcium in neurons increases remarkably after cardiopulmonary-cerebral resuscitation in rats for two hours.There is no effects of propofol on increasing concentration of intracellular free-calcium.
