胶质细胞参与电针足三里对胃运动的调节作用

摘　　要：目的采用胶质细胞代谢阻断剂丙戊茶碱(propentofylline,PPF)予以干预,研究胶质细胞是否参与针刺足三里对胃运动功能调节作用的影响,探讨针刺对胃运动调节作用的神经机制。方法采用免疫荧光组织化学方法,观察大鼠中枢核团迷走复合体(DVC)中神经元被激活的标志c-fos以及星形胶质细胞(GFAP)、小胶质细胞OX42的表达变化。采用胶质细胞阻断剂丙戊茶碱予以干预,观察上述指标的变化。并同步测定大鼠胃电活动的变化。结果对照组大鼠DVC的c-fos呈现低水平表达,GFAP和OX42活化程度很低,胃电亦较为规律。电针足三里组大鼠DVC的c-fos表达水平、GFAP和OX42活化程度明显增加,胃电的波幅和频率均有显著增加。丙戊茶碱干预后电针足三里组大鼠DVC的c-fos表达水平、GFAP和OX42活化程度明显降低,胃电的波幅和频率也有显著的降低,与无干预组相比有显著性差异(P<0.05),与对照组相比也差异显著。结论电针足三里对胃运功的调节不但有神经元的作用,而且胶质细胞也参与这种调节作用。　Objective To investigate whether the glials participate in electro-acupuncture（EA） at Tsusanli regulating gastric activity with propentofylline（PPF） blocking the metabolic function of glials,and to explore neural mechanism of EA at Tsusanli regulating gastric activity.Methods The rats were randomly divided into 4 groups： control group,EA group,NS＋EA group,PPF＋EA group.Using immunofluorescent staining method and electrophysiological method,expression of c-fos,GFAP and OX42 of DVC were determined after EA at Tsusanli in rats with or without interference via cavitas subarachnoidealis.Electrogastrograph（EGG） was recorded synchronously in rats.Results Expression of c-fos,GFAP and OX42 in DVC were lower in control group,and EGG was regular.But their expression in DVC and the amplitude and frequency of EGG increased significantly in EA group.Compared with EA group,expression of c-fos,GFAP and OX42 in DVC and the amplitude and frequency of EGG decreased significantly in PPF＋EA group,but still higher than in control group.There was no significant difference between EA group and NS＋EA group.Conclusion Glias of the dorsal vagal complex may participate in electro-acupuncture of Tsusanli regulating gastric activity.
