摘  要:目的研究ADM及其相关受体CRLR在正常、不孕妇女子宫内膜中的表达,探讨其与不孕的相关关系。方法收集14例原因不明不孕妇女与13例正常生育期妇女黄体中期(LH+6-LH+8)子宫内膜,通过免疫组化、半定量RT-PCR及实时荧光定量RT-PCR,比较两组内膜中ADM、CRLR蛋白表达和基因表达的差异。结果ADM蛋白表达出现在正常内膜的各个细胞中,而在不孕妇女内膜中间质细胞中表达低于正常对照组(P<0.05);CRLR蛋白表达则以血管内皮细胞表达为主,两组内膜各细胞中的表达无差异(P>0.05);不孕妇女内膜中血管密度值为4.81±0.25,明显低于正常妇女的7.83±0.79(P<0.05);ADM在基因水平的表达不孕组ADM mRNA表达3.99倍高于正常组,而CRLR mRNA表达两组无明显差异。结论ADM及其受体CRLR在正常子宫内膜的生成、修复中发挥重要作用;高表达的ADM mRNA尽管有助于种植窗口期孕卵的着床,但其对内膜血管的低调控可能是造成不孕的原因之一。Objective To analyze the different expressions of ADM and CRLR in endometrium of fertile and infertile women and to investigate the correlation between expressions of ADM and CRLR and infertile. Methods The endometrium from 14 unexpected infertile women and 13 healthy fertile women in luteal phase was studied with Immunohistrochemistry, RT- PCR and quantitative RT- PCR. The different expression of ADM and CRLR in protein and mRNA were analyzed. Results ADM showed in all cells in fertile group while less staining was showed in mesenchymocyte in infertile group ( P 〈0.05 ). The staining of CRLR was appeared mainly in epithelial cell with no different staining in two groups. After CD31 staining, the vessel staining in infertile group was lower than that in fertile group (4.81 ±0.25 vs 7.83 ±0179, P 〈0.05 ). The expression of ADM mRNA in infertility was 3.99 times higher than that in fertility while the expression of CRLR mRNA was same in two groups. Conclusion ADM and CRLR play key role in cells growth, differentiation and recovery. The higher expression of ADM mRNA maybe benefit to the implantation during the period of implantation window, however, the lower regulation to the lower vessel density is probably responsible for the unexpected infertility.
