摘  要:目的通过在体外对BXSB狼疮鼠骨髓细胞进行培养,检测BXSB狼疮鼠骨髓细胞的凋亡情况,从而在干细胞水平上探讨系统性红斑狼疮(SLE)的发病机制。方法取4月龄雄性BXSB狼疮鼠和雄性BALB/c小鼠各6只。从小鼠股骨提取骨髓细胞后,用台盼蓝染色法检测骨髓细胞的活性,计数。然后将骨髓细胞悬液加入6孔培养板,每孔加入等量的骨髓细胞。置于37℃、5%CO2中培养24h后收集骨髓细胞,用TUNEL法检测细胞的凋亡。结果①BXSB狼疮鼠骨髓细胞数量明显高于同龄BALB/c小鼠,而且BXSB狼疮鼠与同龄BALB/c小鼠相比,骨髓细胞数/小鼠体重之比也明显升高。②BXSB狼疮鼠骨髓细胞中凋亡水平明显下降。结论BXSB狼疮鼠的发病可能与其骨髓细胞的凋亡异常有关。Objective In order to study a plausible link between SLE and apoptosis in bone marrow cells,we cultured the bone marrow cell of BXSB mice in vitro, and determined the percentage apoptosis in bone marrow cells using the terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeled (TUNEL) method. Methods We used 6 four-month-old male BXSB mice and 6 age-matched male BALB/c mice. Bone marrow cells were isolated from the femurs of mice,and assayed for total cell count and viability using trypan blue dye exclusion. Bone marrow cells were divided into various groups with an equivalent number. Cell samples were cultured for 24h at 37"C, 5 % CO2. After 24h, we gathered the cell samples and detected the percentage apoptosis in bone marrow cells using TUNEL method. Results ①The bone marrow of male BXSB mice contained significantly more cells than age-matched male BALB/c mice did. And expression of cell number on the basis of body weight indicated a trend of increased bone marrow cells for BXSB mice.② Bone marrow cells isolated from BXSB mice display a lower basal level of apoptosis compared to bone marrow cells of BALB/c mice. Conclusion Our study suggested a plausible link between SLE and apoptosis in bone marrow cells.
