摘  要:以硅胶为基质,经表面改性和单克隆抗体固定化,制备了高效液相亲和层析介质。在改性反应中,以γ-缩甘油氧基丙基三甲氧基硅烷为改性剂,比较了水相法键合与有机相法键合的硅烷化率。用合成的α干扰素单克隆抗体高效液相亲和介质纯化由大肠杆菌表达的基因重组人α干扰素,层析过程的活性回收率为94%,纯化倍数为79倍,比活为7.94×106IU/mg。The preparation of high performance liquid affinity chromatographic adsorbents was carried out using γ- glycidoxypropyltrimethoxysilane as reactant to cover the surface of silica gel in aqueous solution. A comparison was made between the reactions in organic phase and in aqueous phase.Anti-interferon-α monoclonal antibody was immobilized onto the prepered adsorbent to form HPLAC medium. Human recombinant 1FN-α expressed in E. coli was purified with the column packed with the prepared adsorbent. The activity recovery was 94% and the purification factor was about 79. The final specific activity of lFN-a was 7. 94 × 106 IU/mg.
