摘 要:目的:研究水飞蓟素脂质体(L-SIL)对卡介苗(BCG)+脂多糖(LPS)引起的小鼠免疫性肝损伤的保护作用。方法:昆明种小鼠80只随机分为空白组、模型组及L-SIL低、中、高剂量组和益肝灵(T-SIL)低、中、高剂量组共8组,测量并比较各组小鼠血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)水平和肝组织匀浆中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)水平以及丙二醛(MDA)、一氧化氮(NO)含量,观察肝脏、脾脏系数的变化,并观察各组小鼠肝组织病理学改变。结果:与模型组相比,L-SIL各剂量组血清ALT、AST水平明显降低(P〈0.05-0.01),肝组织匀浆GSH-PX、SOD水平明显升高(P〈0.05-0.01),MDA和NO含量明显降低(P〈0.01),肝脏、脾脏系数明显降低(P〈0.05-0.01)。L-SIL对ALT、AST、GSH-PX水平和NO含量的作用强于同剂量T-SIL组(P〈0.05-0.01)。组织学观察L-SIL低、中剂量组的肝损伤明显轻于模型组和相应剂量T-SIL组。结论:L-SIL对小鼠BCG+LPS引起的免疫性肝损伤有明显的保护作用,且其保护作用明显优于T-SIL。Objective: To investigate the protective effect of silymarin liposomes (L-SIL) on the BCG+LPS- induced liver injury in mice compared with silymarin tablets (T-SIL). Methods: The models of hepatic damage were induced by BCG+ LPS. The alanine aminotransferase (ALT), aspartate aminotransferase (AST) activities in serum, malondialdehyde (MDA), nitric oxide (NO) contents, superoxide dismutase (SOD), glutathione peroxidase (GSH-PX) levels in liver homogenate, coefficient of liver were examined and dimensions of hepatic damage in the histopathology were observed. Results: L-SIL could inhibit the rising of serum ALT, AST activities caused by BCG+LPS. Meanwhile, L-SIL also decreased MDA, NO contents, reduced the coefficient of liver and prevented the reduction of SOD, GSH-PX activities in the liver homogenate. The effects of L-SIL on the AI.T, AST activity in serum,GSH-PX activities and NO content in liver homogenate against experimental liver injury in mice were more powerful than that of T-SIL of the same dose. The liver pathological observation showed that the liver injuries of L-SIL (75,150 mg · kg^-1 · d^-1) were significantly less than that of the model group and T-SIL of the same dose. Conclusion: The protective effect of L-SIL on the BCG+LPS-induced liver injury in mice was more potent than that of the same dose of T SII.
