循环型和局部型抗精子抗体的差异研究

摘　　要：目的探讨免疫性不孕患者体内抗精子抗体循环型与局部型识别免疫性不孕相关精子抗原的差别。方法以二维电泳技术分离正常精子总蛋白,比较免疫性不孕患者的血清和宫颈黏液识别精子抗原的差别。结果蛋白印迹试验显示免疫性不孕患者及正常生育力妇女的血清均能识别精子蛋白;二维电泳技术结合免疫印迹试验发现免疫性不孕患者体内抗精子抗体循环型可识别约200余种抗原,而局部型识别的精子抗原约100余种,且分布的图谱明显不同;资料的回顾比较性分析显示局部型抗体能识别更多的精子膜抗原。结论抗精子抗体的不同类型在免疫性不孕的诊治中具有不同的意义。女性宫颈黏液抗精子抗体能识别更多的不孕相关精子膜抗原,对其的深入研究将有助于免疫性不孕相关精子抗原的筛选和鉴定。Objective To evaluate the difference of immue infertility associated antigen reconginized by serum and cervical mucus. Methods Two-dimentional electrophoresis and western blot analyses were used to identify the difference with different kinds of ASA from immune infertility women. Results All samples from patient could react with total sperm extracts,as well as the normal women （2/4 cases）. The sera from inferitlity could recognize about 200 kinds of spots by 2-D plus western blot analyse. Similarly,for the cervical mucus,it was about 100. However, the patterns of protein spots were different. The retrospective analysise shown that local ASA could react with sperm membrane antigen more than sera. Conclusion Different kinds of ASA could react with different about immunologically characterized sperm proteins. It will conduce to identify and characterize the iditope immunoreactive sperm antigens with cervical mucus from immune infertility women.
