摘  要:目的:探讨莪术提取物榄香烯抑制神经胶质瘤细胞体外增殖的机制及其对神经胶质瘤细胞体内增殖的影响。方法:采用细胞计数、Western印迹等方法分别检测不同浓度和不同作用时间榄香烯对胶质瘤细胞的增殖影响和对ERK、Akt蛋白质表达的影响,并观察接种胶质瘤细胞裸鼠受榄香烯作用后肿瘤增殖的变化。结果:榄香烯对大鼠C6胶质瘤细胞具有明显的抑制增殖作用,该增殖抑制效应呈剂量、时间依赖性。榄香烯可明显下调磷酸化ERK表达,同样呈剂量、时间依赖性,而对Akt表达无明显影响。榄香烯可明显抑制C6胶质瘤细胞的体内增殖。结论:榄香烯能有效抑制胶质瘤细胞的体内和体外增殖,下调磷酸化ERK蛋白质表达可能是榄香烯抑制C6细胞增殖的机制。Objective: To study the mechanism underlying the anti-proliferation effect of elemene, an extract of Zedoary Turmeric Oil, on rat C6 glioblastoma cells in vitro and observe the influence of elemene on the proliferation of C6 glioblastoma cells in vivo. Methods:Effect of elemene on the proliferation of C6 glioblastoma cells and expression of extracellular signal-regulated kinase (ERK) and Akt protein was measured by cell-counting and Western blotting analysis at different concentrations and at different time points, respectively. Development of solid tumor growth was observed after C6 glioblastoma cells were inoculated in nude mice. Results:Elemene had significant inhibitory effects on the proliferation of C6 glioblastoma cells in a dose- and time-dependent manner. Elemene down-regulated the expression of phosphorylated ERK dose- and time-dependently but had no significant influence on the expression of Akt protein. Elemene significantly suppressed the growth of C6 glioblastoma cells in nude mice. Conclusion:Elemene effectively inhibited growth of C6 glioblastoma cells in vivo and in vitro through down-regulation of phosphorylated ERK.
